Objective:Observe the effect of acupuncture on the antioxidants in the brain tissue of VCI rats,analyze whether acupuncture can regulate the Nrf2-ARE signal pathway,thereby protecting the nerve cells of VCI rats,and provide a theoretical basis for acupuncture treatment of VCI.Methods:Experiment 1:The effect of acupuncture on the behavior and hippocampus morphology of VCI ratsSixty-four male SD rats were selected and randomly divided into sham operation group,model group,scalp acupuncture group,and nape acupuncture group.The sham operation group only exposed bilateral carotid arteries.VCI rat models were prepared in the model group,and the scalp acupuncture group was in Scalp acupuncture was given on the basis of successful model preparation,and the nape acupuncture group was given nape acupuncture as an intervention method based on the successful model preparation.Morris water maze behavior experiment was used as the behavior detection index;Nissl staining was used to analyze the morphological changes.Experiment 2:The effect of acupuncture on the expression of related factors in the Nrf2-ARE signaling pathwayEighty female SD rats were randomly divided into sham operation group,model group and scalp acupuncture group,nape acupuncture group,scalp acupuncture+nape acupuncture group.Only the carotid artery was exposed in the sham operation group;the rat VCI model was prepared in the model group;the scalp acupuncture group was given scalp acupuncture on the basis of successful model preparation,and the nape acupuncture group was given nape acupuncture on the basis of successful model preparation,scalp acupuncture+Nape acupuncture group was treated with scalp acupuncture+nape acupuncture group on the basis of successful model preparation;transmission electron microscopy scanning method was used to analyze the ultrastructure of rat hippocampal neurons;Elsia method was used to detect rat MDA,GSH,SOD content;Western blot detection was great Mouse hippocampal CytC,cleaved Caspase-3,Bax,Bcl-2,Nrf2,HO-1 and NQO1 protein;RT-PCR method was used to detect the expression of mouse hippocampus Nrf2mRNA,HO-1mRNA and NQO1mRNA.Result:1.Comparison of water maze test scores:the escape times and latency of the model group were significantly different from that of the sham operation group(P<0.01),indicating the success of modeling.Compared with the model group,the number of escape and latency in acupuncture group were significantly different(P<0.01).Compared with other acupuncture groups,there were significant differences in the escape times and latency of acupuncture group(P<0.01).There was no significant difference between the head acupuncture group and the neck acupuncture group in the number of escape and the latency(P>0.05).2.HE staining results:The hippocampal neurons of rats in the sham operation group had complete structure,clear hierarchy,regular morphology,compact and orderly arrangement,uniform distribution,large and regular nuclei,and no obvious loss of neurons.In the model group,the hierarchy of neurons in the hippocampus was poor,with irregular morphology,loose and disordered arrangement,intercellular edema,deep pyknosis of nuclei,enlarged and wider space between neurons and surrounding tissues,and obvious loss of neurons.In the head acupuncture group and neck acupuncture group,the structure of the neurons in the hippocampus was poor,with irregular arrangement,intercellular edema,nuclear pyknosis,low hyperchromaticity,smaller gap between neurons and surrounding tissues,and loss of neurons.The neuronal structure of hippocampal tissue of rats in the head acupuncture+nangeal acupuncture group was relatively complete and arranged evenly,the intercellular edema was significantly reduced,the nuclear solid shrinkage was less,the degree of hyperchromaticity was reduced,the gap between neurons and surrounding tissues was smaller,and a few neurons were lost.3.Elsia results:Compared with sham operation group,the expression of MDA in the hippocampus of model group and acupuncture group was significantly increased(P<0.01),while the expression of GSH and SOD were significantly decreased(P<0.01).Compared with the model group,the expression of MDA in the hippocampus of acupuncture group decreased(P<0.01),and the expression of GSH and SOD increased significantly(P<0.01).Compared with other acupuncture groups,the expression of MDA and GSH and SOD in the hippocampus of acupuncture group decreased significantly(P<0.01).Compared with the head acupuncture group,the expression levels of various substances in the hippocampus of the neck acupuncture group were similar,but there was no significant difference(P>0.05).4.Western blot test results:Compared with the sham operation group,the expression of CytC,cleaved caspase-3 and Bax in the hippocampus of the model group was significantly increased(P<0.01),while the expression of Bcl-2 was decreased(P<0.01).Compared with model group,the expression of CytC,cleaved caspase-3 and Bax in hippocampus of acupuncture group was significantly decreased(P<0.01),and the expression of Bcl-2 was significantly increased(P<0.01).Compared with head acupuncture group and neck acupuncture group,the expressions of CytC,cleaved caspase-3 and Bax in the hippocampus of head acupuncture+neck acupuncture group were decreased(P<0.01),and the expression of Bcl-2 was significantly increased(P<0.01).Compared with the head acupuncture group,the expression levels of various substances in the hippocampus of the neck acupuncture group were similar,but there was no significant difference(P>0.05);Compared with sham operation group,the expressions of Nrf2,HO-1 and NQO1 in hippocampus of model group were significantly decreased(P<0.01).Compared with model group,the expressions of Nrf2,HO-1 and NQO1 in hippocampus of acupuncture group were significantly increased(P<0.01)).The expressions of Nrf2,HO-1 and NQO1 in the hippocampal tissues of rats in the head-acupuncture+head-acupuncture group were increased compared with those in the head-acupuncture+head-acupuncture group(P<0.01).Compared with the head acupuncture group,the expression levels of Nrf2,HO-1 and NQO1 in the hippocampus of the neck acupuncture group were similar,and there was no significant difference between the two groups(P>0.05).5.RT-PCR test results:The expressions of Nrf2mRNA,HO-1mRNA,and NQO1mRNA in the model group and the acupuncture group were significantly higher than those in the sham operation group(P<0.01);the expressions of Nrf2mRNA,HO-1mRNA,and NQO1mRNA in the acupuncture group were significantly higher than those in the model group(P<0.01);The expression of Nrf2mRNA,HO-1mRNA,and NQO1mRNA in the scalp acupuncture+nape acupuncture group was significantly higher than that of other acupuncture groups(P<0.01);the expression of Nrf2mRNA,HO-1Mrna,and NQO1mRNA in the nape acupuncture group was similar to that of the scalp acupuncture group,No significant difference(P>0.05).Conclusion:1.Acupuncture can improve the cognitive function of VCI rats and reduce the neuronal damage in the hippocampus of VCI rats,and the treatment effect of head acupuncture combined with neck acupuncture is the best.2.Acupuncture can promote the anti-oxidative stress response in the hippocampus of VCI rats,increase the expression of antioxidants,and reduce the synthesis of MDA.3.Acupuncture can inhibit the expression of Cytc,caspase-3,Bax,increase the expression of BCL-2,and reduce the production of cell apoptosis.4.Acupuncture may regulate the Nrf2-ARE signaling pathway to increase the synthesis of downstream HO-1 and NQO1,activate the production of antioxidants GSH and SOD,reduce the content of lipid peroxide MDA,and reduce apoptosis Produced,thereby protecting the hippocampus tissue of the sea and improving the cognitive function of rats.
