Purpose The pathogenesis of myopia has not been fully clarified.Non-coding RNA may be involved in the pathogenesis of high myopia.In this study,we aim to observe the characteristics of the anterior segment in highly myopic eyes;to detect and screen differentially expressed micro RNA in high myopia from clinical samples,and to study their roles in myopiaMethods1.Characteristics of anterior segment in highly myopic eyes were observed and the influence of implantable collamer lens(ICL)implantation on ocular anterior segment was analyzed.2.Aqueous humor from ICL implantation and cataract surgeries was collected for micro RNA next-generation sequencing.Differentially expressed micro RNAs were screened for functional and pathway analysis.3.The expression levels of mi R-29 a in plasma and aqueous humor of patients with high myopia was detected.4.Human scleral fibroblasts were isolated and cultured in vitro.After transfection with mi R-29 a mimics and inhibitors,the protein expression changes were detected by i TRAQ protein profiling;and collagen synthesis,cell proliferation,cell migration of scleral fibroblasts were detected.Results1.Anterior chamber volume can better monitor the anterior segment of highly myopic eyes than traditional parameters.ICL implantation results in pupil diameter reduction under low and medium illuminance conditions.ICL central hole design has significant influence on the position of anterior segment structures during iris or ciliary muscle movement.2.The expression levels of mi R-889,mi R-369,mi R-370,mi R-495,mi R-127 in the aqueous humor of highly myopic eyes were significantly increased;mi R-10 b,mir-143,mir-145 significantly decreased.Analisis of target genes and regulatory pathways of these micro RNAs show possible involvement in the pathogenesis of high myopia.The expression levels of mi R-214,mi R-103,mi R-98,let-7 family,mir-29 family were significantly increased in the aqueous humor from highly myopic eyes.3.Mir-29 a expression levels in aqueous humor and plasma of patients with high myopia were significantly increased.4.In vitro cultured human scleral fibroblasts,mir-29 a significantly inhibited the expression level of collagen type I and enhanced the cell migration ability,but had no significant effect on cell proliferationConclusions Anterior chamber volume is an important parameter,especially for highly myopic eyes.ICL implantation has significant influence on pupil diameter,light reflex,and position of anterior segment structures during iris or ciliary muscle movement.Aqueous humor of highly myopic eyes shows a specific micro RNA expression profile;differentially expressed micro RNAs may be involved in the pathogenesis of high myopia.Mi R-29 a has the potential to be a biomarker and new therapeutic target of high myopia.Mi R-29 a may be involved in sclera remodeling process through modulating collagen synthesis in scleral fibroblasts.
