Objective: To analyze the clinical value of MCM family gene for lower grade gliomas by studying the biological function and metabolic pathway of MCM family gene.The expression of MCM2 and MCM3 in LGG was confirmed by single center cohort study.Materials and methods: LGG-related m RNA sequencing data sets of MCM gene family and clinical data were downloaded from TCGA database.Analysis of the clinical value of MCM family genes for lower grade brain gliomas by the study of comparison of MCM gene expression in normal brain tissue and brain tumor tissue,comparison of MCM gene expression in different grades of LGG.To establish a nomogram model to predict the prognosis of LGG and analyze the related MCM family genes and clinical related factors.GSEA was used to analyze the accumulation of genes and their coexpression genes at transcriptome level which had the greatest influence on LGG prognosis.To analyze the biological process and pathway of these genes,and to explore the mechanism of MCM family genes in the development of LGG.A retrospective study was conducted on 75 LGG patients from May 2013 to December 2018 in the Guangxi Medical University Affiliated Tumor Hospital and the clinical data was collected.Analysis the results of immunohistochemistry,the Kaplan-Meier survival curve and multi factor Cox risk model were used to analyze the clinical factors and prognosis.Results: The expression of MCM2-7 in LGG was significantly higher than that in normal brain tissue,and the expression of MCM2-7 in WHO-III LGG was significantly higher than that in WHO-II LGG.MCM2,MCM3 and MCM4 were correlated with the overall survival time of LGG.MCM2 was associated with progression free survival of LGG.The nomogram model suggested that the overall survival time and progression free survival time of LGG patients who were over 65 years old and with high expression level of MCM2 gene were poor.MCM2,MCM3 and MCM4 genes are involved in DNA replication,cell cycle and p53 regulation,which affect the prognosis of LGG patients.In the cohort study,MCM3 protein expression was correlated with overall survival time and progression free survival time of LGG patients,the prognosis of patients with positive MCM3 expression was worse than that of negative.Conclusion: The family of MCM proteins may participate in many biological processes and pathways to affect the prognosis of LGG patients.MCM2 and MCM3 proteins are new biomarkers related to the prognosis of lower grade gliomas.This result needs to be further verified in the future.
