Background: Acute kidney injury(AKI)is a common clinical complication associated with high mortality in patients.Immune cells and cytokines have been described recently as playing essential roles in AKI pathogenesis.Plasmacytoid dendritic cells(pDCs)are a unique DC subset that specializes in type I interferons(IFNs)production.pDCs have powerful functionalities to orchestrate innate and adaptive immune responses.However,to date the characterization of the potential role of pDCs in AKI remains unknown.Object:1.To observe the kinetics of pDCs and its phenotype in kidney after AKI.2.To detect pDCs function in AKI by deletion of pDCs.3.Analysis the role of IFN-? in the function of pDCs during AKI.4.To analysis the cellular and molecular mechanisms in pDCs recruitment and activation.5.The function of pDCs in another murine model of AKI Results:1.pDCs rapidly infiltrates into kidney and activates in response to AKI.2.Deletion of pDCs protected mice from cisplatin induced AKI,with reduced proinflammatory cytokine production,inhibited immune cells infiltration and apoptosis in kidney.3.pDCs contributes to kidney damage by the IFN-? pathway.4.Renal tubular epithelial cells(TECs)may facilitate pDCs recruitment and activation by regulating cytokines and chemokines production in AKI microenvironment.5.Similar function of pDCs were observed in ischemia reperfusion injury induced murine model of AKI.Conclusion: pDCs play detrimental roles in AKI by production IFN-?.
