Aims: Micro RNAs(miRNAs)are a class of single-stranded non-coding RNAs that post-transcriptionally regulate gene expression with the target m RNA by specific base pairing,including negative regulation and positive regulation,while,the mechanism of positive regulation is unclear.The regulatory effect of miRNA includ cell apoptosis,growth,differentiation of hematopoietic cells,fat metabolism process of life.Cervical cancer is one of the most common gynecologic malignant tumor,its incidence was younger trend in recent years,a serious threat to women's health.Tumor development is a multi factor complex,multistep process,cervical cancer is no exception.In recent years,also found that miRNA in cervical cancer tissue and serum differentially expression,miRNA may be involved in the progression of cervical cancer.The thesis mainly studies the micro RNA-27 a and its target gene H2.0-like homeobox(HLX)play in the role of cervical cancer cells,and the mechanism research.Methods: We first use MTT,plate colony formation assay,invasion and migration of Transwell experimental study effcefs of miR-27 a in cervical cancer cells He La.After the biological information web site predicted that the miR-27 a of several potential target genes,selected one target gene HLX associated with tumor growth as the research object.Using EGFP fluorescent reporter vector experiment,qRT-PCR and Western blot technology to verify the miR-27 a of target gene HLX in the role of direct and regulation.Also use MTT,plate colony formation assay,invasion and migration of Transwell experimental study effcefs HLX in cervical cancer cells He La.Finally through MTT,plate colony formation assay,invasion and migration of Transwell experiment to save the effect,further proved miR-27 a affect malignant behavior of cervical cancer cells through its target gene HLX.Results: Overexpression of miR-27 a promotes growth,migration and invasion of He La cell.Using the bioinformatics prediction of target genes,HLX is a candidate target gene of miR-27 a,The HLX m RNA 3'-untranslated region(3'UTR)contains the potential binding site of miR-27 a.The EGFP fluorescent report system also confirmed that miR-27 a can directly bind to the HLX m RNA 3'UTR and positively regulate the gene expression.qRT-PCR and Western blot was further validated miR-27 a upregulation of endogenous HLX expression.The study showed that over expression of target gene HLX function,HLX also promotes growth,migration and invasion of He La cell.Conclusions: We show that in cervical carcinoma cell He La cell,miR-27 a through the up regulation of its target gene HLX,affect cervical cancer cell growth,migration and invasion...
