Study On The Effect Of Jingning Granules On Children With Qi-Yin Deficiency Syndrome Of Attention Deficit Hyperactivity Disorder And SHR Behavior

Objective:To explore the behavior changes after treatment of Jing ning granule in spontaneously hypertensive rats.To explore the clinical efficacy and adverse reactions of Jing ning Granule in the treatment of children with ADHD?Methods:In animal experiments,2-week-old spontaneously hypertensive rats were chosen as model rats of children's attention deficit hyperactivity disorder disease,half male and half fe male,with WKY rats as control mice.Professor Wang Jun Hong's experien ced prescription-Jing ning particles was used as intervention drug in our experiments and the particles of fi nished product were provided by Professor Ni Jian's research group of traditional Chinese medicine Academy of Beijing University of Chinese Medicine University.Pediatric Huang long granule,Methylphenidate were selected as a control drug.The total intervention time was 2 weeks,after 2 weeks intervention,the opening experiment and social behavior experiment recorded the Distance moved Total?Velocity?Moving/Center-point Cumulative Duration?Body elongation Normal Cumulative Duration?Stranger side(center-point,s)?Empty side Cumulative Duration(center-point,s)?Stranger-sniffing zone Cumulative Duration(nose-point,s)?Empty-sniffing zone Cumulative Duration(center-point,s)?S-S/C(Stranger-Sniffing/Chamber)?Distance moved(center-point,cm).These indexes are to reflect the autonomic activities and social activities of the rats.In clinical experiments,during September 2013 to October 2015,children with attention deficit hyperactivity disorder were selected from the pediatric clinic of Beijing University of traditional Chinese medicine Dong zhi men Hospital.the pediatric clinic of Beijing He ping li hospital and primary schools of Dong Cheng District.The traditional Chinese medicine and Western medicine diagnostic criterias were incorporated,and informed consent was signed.After taking Jing ning formula granule orally for 3 months,some indexes of the two groups before and after treatment were measured and recorded,such as neurological soft signs,TCM syndrome score,behavior science scale score,Conners parent symptom questionnaire score,cancellation test,blood,urine,stool routine examination,liver and kidney(ALT,BUN,Cr)function test,ECG examination.Adverse events were recorded.The corresponding evaluation criteria was established.Results:The Distance moved Total and Velocity of spontaneously hypertensive rats in the open field test,the control group(WKY group)were significantly different with other groups(*P<0.05),after 2 weeks treatment,Methylphenidate group,pediatric Huang long granule group and model group were statistically significant difference(**P<0.05),Jing ning particles group's Velocity decreased more obviously.For the Moving/Center-point Cumulative Duration and Body elongation Normal Cumulative Duration,the control group(WKY group)were significantly different with other groups(*P<0.05),after 2 weeks treatment,Methylphenidate group,pediatric Huang long granule group and model group had no statistically significant difference(P>0.05),but Jing ning particles group' s Moving/Center-point Cumulative Duration had significant difference with model group(*P<0.05).In the social behavior experiment,the Stranger side(center-point,s)and the Empty side Cumulative Duration(center-point,s),the control group(WKY group)were significantly different with other groups(*P<0.05),after 2 weeks treatment,Methylphenidate group,pediatric Huang long granule group and model group had no statistically significant difference(P>0.05),Jing ning particles group also had no statistically difference(P>0.05).The Stranger-sniffing zone Cumulative Duration(nose-point,s)and the Empty-sniffing zone Cumulative Duration(center-point,s),the control group(WKY group)were significantly different with other groups(*P<0.05),after 2 weeks treatment,Methylphenidate group,pediatric Huang long granule group and model group had no statistically significant difference(P>0.05).The S-S/C(Stranger-Sniffing/Chamber),the control group(WKY group)were significantly different with other groups(*P<0.05),after 2 weeks treatment,Methylphenidate group,pediatric Huang long granule group and model group had statistically significant difference(**P<0.05),Jing ning particles group was more significant different than pediatric Huang long granule group and Ritalin group.The Distance moved(center-point),the control group(WKY group)were significantly different with other groups(*P<0.05),after 2 weeks treatment,Methylphenidate group,pediatric Huang long granule group and model group had no statistically significant difference(P>0.05),Jing ning particles group also had no significant improvement.In clinical experiment,after 3 months treatment,there were 14 cases markedly effective,markedly effective rate is 25%,36 cases effective,effective rate is 64.29%,6 cases invalid,invalid rate is 10.74%,so 50 cases are total effective,and the total efficiency is 89.28%.Soft neurological signs were improved,the total score of SNAP-? was improved,there were significant differences(P<0.05).The total score of TCM symptoms improved,there were significant differences(P<0.05).The main TCM syndrome total score was improved,there was significant difference in Statistics(P<0.05).The main symptoms of traditional Chinese medicine(hyperactivity restless,inattention,low learning efficiency)were statistically significant differences before and after treatment(P<0.01);secondary symptoms(insomnia and dreaminess,hot feeling in hands and feet,dry mouth,lusterless face and spontaneous sweating,night sweats)also had significant difference before and after treatment(P<0.05).Behavior Scale total score was improved,there was significant difference in Statistics(P<0.05).The hyperactivity index was improved,there are significant differences in Statistics(P<0.05).The score and leakage cancellation number of strokes in the cancellation experiment had statistical difference(P<0.01),wrong number did not have statistically difference(P>0.05).Conners parent scale scores was improved,there was significant difference in Statistics(P<0.05).In the blood,urine,stool routine tests,liver and kidney(ALT,BUN,Cr)functional tests,electrocardiogram test,no obviously abnormal were found because of the use of Jing ning particles,in the analysis of adverse events,no adverse events were found by taking in Jing ning granules.Conclusion:1.Jing ning Granule can improve the autonomic activity of spontaneously hypertensive rats.2.Jing ning Granule can improve the social activities of the spontaneously hypertensive rats.3.Jing ning Granule has a good improvement on the symptoms of children with attention-deficit hyperactivity disorder.4.Jing ning Granule is generally safe in intervention of children with ADHD.
