| Objective:Through the study of KLF4 and downstream gene expression in rat gastric mucosa,we explored the mechanism of the effect of Xin Wei Granule on LGIN,and evaluated the therapeutic effect of Xin Wei Granule on LGIN in rats.Methods:110 male Wistar rats were randomly divided into two groups:model group 80,blank group 30,MNNG composite method was used in preparing the model in 16 weeks.During the modeling period,the model group rats can drink MNNG solution freely,take the standard diet containing ranitidine,receive gavage of hot Sodium Chloride Solution,and be hunger and full without regular.To observe the pathological changes of gastric mucosa in rats,after confirming the models in accordance with the LGIN criteria,stop continuing to build the model,and step into the treatment phase.60 rats selected randomly from the remaining rats of model group were divided into four groups:model control group(treated with 0.9%sodium chloride solution),western medicine control group(treated with dimensional enzyme fibroin solution),Chinese medicine control group(treated with WeiFuchun solution),XinWei granule treatment group(treated with Xinwei granules solution).15 rats in each group were given 1 time gavage daily;15 rats selected randomly from blank group were given regular feeding,lasting for 12 weeks.To observe the general state of rats in each group,gross morphology of gastric mucosa,pathological changes of gastric mucosa,and detection of KLF4,p21WAF1/CIP1、cyclinD1 and protein expression in gastric mucosal tissues by real-time PCR and Western blot methods were applied.Result:1.The general state of the model animals and the gross morphology and histopathology were in accordance with the characteristics of LGIN model.After treatment,the pathological changes of each drug treatment group were improved,and the improvement of the XinWei granule treatment group and the WeiFuchun treatment group was obvious.2.The results of Western blot and Real-time PCR show that:Compared with the blank control group,the expression levels of KLF4,p21WAF1/CIP1 in the model group and the treatment group rats gastric mucosa was significantly decreased,P<0.05.But compared with the model group,the expression level of KLF4、p21WAF1/CIP1 in all treatment groups rats gastric mucosa,increased significantly,P<0.05.There was no significant difference between the Xin Wei granule group and the WeiFuchun group,P>0.05.And compared with the enzyme group,the expression level of KLF4,p21WAF1/CIP1 in the gastric mucosal tissue of the two traditional Chinese medicine treatment group was increased,P<0.05,and the difference was statistically significant.3.The results of Western blot and Real-time PCR show that:Compared with the blank control group,the expression levels of CyclinD1 in the model group and the treatment group rats gastric mucosa was significantly increased,P<0.05.But compared with the model group,the expression level of CyclinDl in all treatment groups rats gastric mucosa,decreased significantly,P<0.05.There was no significant difference between the Xin Wei granule group and the WeiFuchun group,P>0.05.And compared with the enzyme group,the expression level of CyclinDl in the gastric mucosal tissue of the two traditional Chinese medicine treatment group was decreased,P<0.05,the difference was statistically significant.Conclusion:1.Xin Wei granule can effectively improve the pathological changes of gastric mucosa in the gastric mucosa of the Low grade intraepithelial neoplasia of the gastric mucosa.2.Xin Wei granule can increase the expression of KLF4 in the gastric mucosa of the Low grade intraepithelial neoplasia of the gastric mucosa,up-regulate its downstream tumor suppressor gene p21WAF1/CIP1,inhibit the expression of proto-oncogene CyclinD1,which may be the important mechanism of the Xin Wei granule to reverse the gastric mucosal low grade intraepithelial neoplasia. |
