Purpose: Diabetic bladder dysfunction (DBD) is a common and bothersomecomplication of both types I and II diabetes mellitus (DM), but the underlyingmechanisms are not fully defined and the therapeutic strategies are still limited. Whilebladder dysfunction has been extensively studied in animal models of type I DM,little is known about DBD associated with type II DM, due to lack of an appropriateanimal model. Therefore, we aimed to characterize the functional and structuralalterations of urinary bladder of db/db obese type II diabetic mouse model on FVBgenetic background.Methods: Male FVB db/db mice were compared with age-matched FVB NJcontrol mice for52weeks old. At6,12,24and52weeks old, body weight, bloodglucose and glycated hemoglobin levels were measured. Bladder functions wereassessed by24hour micturition analysis and conscious cystometry. Masson'sTrichrome staining and morphometric analysis were used for histological evaluation.Results: FVB db/db mice displayed detrusor overactivity, characterized by asignificant increase in frequency of non-voiding contractions during the filling phase,together with an increase in voided volume, bladder capacity, intercontraction intervaland micturition pressure. This was associated with histological alterations in thecomposition of bladder wall components. Conclusion: These data demonstrated that FVB db/db mouse model appears todisplay similar features of structural and functional bladder remodeling, seen indiabetic patients. We suggest that FVB db/db mouse is a suitable model for bladderresearch to further elucidate the mechanistic pathways of type II DM-induced bladderdysfunction.
