Objective:To investigate the protective effects of electro-acupuncture of“jiajixue”on myocardial ischemia and reperfusion injury in rats,endothelial function、antioxidation、the extent of apoptosis in cardiomyocytes ,and the expression of Bcl-2/Bax protein ,to explore the possible protective mechanism of acupuncture of“jiajixue”on myocardial ischemia and reperfusion injury.Methods:50 wistar rats were randomly divided into 5 groups: Jiaji group, Neiguan group, model group, sham-operation group, Yanglingquan group ,with 10 rats in each group. Myocardial ischemia and reperfusion injury model was duplicated by occlusion of the descending anterior branch of the left coronary artery. The content of NO、ET、IL-1β、SOD、MDA in serum were measured by ELISA. Apoptotic myocytes were detected with the TUNEL method. The expression of Bcl-2 and Bax protein in myocardial cells was detected by immmunohistochemistry.Results:1. Compared with the sham-operation, NO content significantly decreased(p﹤0.01), IL-1βcontent significantly increased in the other group(p﹤0.01);ET content significantly increased in model and Yanglingquan groups(p﹤0.01).Compared with the model ,NO content significantly increased in the Neiguan group(p﹤0.05); ET content significantly decreased in the Jiaji group(p﹤0.01); ET content also significantly decreased in the Neiguan group(p﹤0.05); IL-1βcontent significantly decreased in the Jiaji and Neiguan groups(p﹤0.01);NO、ET、IL-1βcontent not significantly changed in Yanglingquan group(p>0.05).Compare Jiaji with Neiguan group,NO and IL-1βcontent of serum not significantly changed(p > 0.05),but the content of ET was significantly decreased in Jiaji group (p﹤0.01).2.Compared with the sham-operation, MDAcontent significantly increased(p﹤0.01),SOD content significantly decreased in model and Yanglingquan groups(p﹤0.01),SOD content significantly decreased in Neiguan group(p﹤0.05). Compared with the model, SOD and MDA content were significantly changed in the Jiaji and Neiguan groups(p﹤0.01),but SOD and MDA content were not significantly changed in Yanglingquan group(p>0.05). Compare Jiaji with Neiguan group,SOD and MDA content of serum were not significantly changed(p>0.05).3.Compared with that in model group, the cardiomyocytes apoptosis was markedly inhibited in Jiaji group, Neiguan group and sham-operation group,Apoptosis index(AI)were significantly decreased((8.846±1.845)%、(7.950±1.671)%、(1.310±0.492)%vs. (18.542±1.894)%,respectively (P﹤0.01). Compared with the sham-operation, Apoptosis index(AI)were significantly increased in other groups (P﹤0.01). Compare Jiaji with Neiguan group, Apoptosis index(AI) were not significantly changed(p>0.05).4 . The result of the expression of Bcl-2 and Bax protein measured by immuno-histochemistry:The expression of Bcl-2 and Bax protein were less in the sham-operation group, The expression of Bcl-2 and Bax protein was significantly increased in model groups compared with that in the sham-operation group(P﹤0.01), while the expression of Bax protein was increased significantly. Compared with the model group , the expression of Bax protein was significantly decreased and the expression of Bcl-2 was significantly increased in Jiaji group and Neiguan group(P﹤0.01). Compare Jiaji with Neiguan group, the expression of Bcl-2 and Bax protein were not significantly changed(p>0.05),but the expression of Bcl-2 and Bax protein were significantly changed (P﹤0.01).Conclusion:Eelectroacupucture of“jiajixue”have effective protective effects on myocardial ischemia and reperfusion injury. The mechanism probably relate to increasing the secretion of NO and the activity of SOD, inhibiting the secretion of ET and contents of IL-1βand MDA ,and inhibiting the cardiomyocytes apoptosis by adjustment of Bcl-2 and Bax gene. |
