To study the effects of Vasonatrin peptide(VNP)on the survival, expressions of NO and iNOS in skin flap crush injury. Methods Forty-eight rats were randomly divided into four groups: the SHAM group, the crush injury group with normal saline treated group, dexamethasone treated group and VNP treated group. Tissue samples from skin flap were collected from 12h, 24h, 48h, 72h after surgery,skin flap survival rate was calculated. The expressions of iNOS in the skin flap was detected by immunohistochemistry staining and NO concentration in the serum was detected by nitric acid reductase. Results: Compared with dexamethasone treated group, VNP could increase the survival rate of skin flap. The NO concentration in serum was almost normal and no discernible fluctuate was detected. The NO concentration was dramatically increased in SHAM group. Dexamethasone significantly decreased the concentration of NO. The NO concentration in VNP treated group was lower than that of dexamethasone. The expression of iNOS was almost negative in SHAM group, and the expression of iNOS in saline treated group was significantly higher than that of SHAM group. Expression of iNOS in dexamethasone treated group was lower than that of saline group and VNP further decreased the iNOS expression. Conclusion: VNP could promote the survival of skin flap in the crush injury through decreasing the expressions of iNOS in skin flap and NO concentration in serum. VNP has the potential to be a new therapeutic drug for clinical application.
