Anisodus luridus var.were perennial herbs of the Solanaceae,Anisodus.This was a traditional Tibetan medicine and can also be used as an important plant source of scopolamine alkaloids,which has great development and utilization value.The study was of great significance of A.luridus.On the one hand,it has certain theoretical significance to explain its adaptation to drought,low temperature,high temperature,strong ultraviolet,etc.;on the other hand,studying the tissue difference of the effective components of A.luridus through metabolome and transcriptome Provide support for the development and utilization of A.luridus.The study was starts with the distribution of this resources in Tibet of A.luridus.The study used the Maxent model to predict the appropriate distribution area.Through the analysis,it was found that the temperature,soil,humidity index and other factors have a greater influence on the distribution of Lingdangzi.Then,The study uesd seeds of A.luridus as the research object to study the response of the seeds stage to different treatments.By processing this seedlings to detect the changes of its physiological level,to study the response mechanism of this seeds at the plant physiological level.Finally,the effects of drought,low temperature,high temperature,ultraviolet and other stress treatments on the transcription and metabolism of A.luridus were tested,and the adaptation mechanism of A.luridus at the molecular level and metabolome level was briefly analyzed.The main results of this study are as follows:1.Field investigations found that was mainly distributed in the southeast and east of A.luridus in Tibet with an altitude of 2942-4410 m.The habitat types are meadows,shrubs,and coniferous forests.The study found that the most important ecological factors affecting the distribution of bells are temperature,soil type and wetness index.2.The biological study of the seeds of A.luridus,This had shown that the storage period of the seeds is more than 5 years,and the germination range of the seeds is 15-30℃.This had good adaptability to low concentration of salt and PEG-6000;The seeds do not have dormancy However,the application of GA3 can increase the germination speed of the seeds;because the seeds of the A.luridus cannot spread farther away,this study selected the litter of the A.luridus to study their allelopathy.The study showed that low concentrations of litter of the seed germination has a promoting effect.On the difficulty of reaching 0.05g/m L,the germination rate of A.luridus was about 25%.3.Studying the changes of physiological indexes of A.luridus during stress treatment,it was found that its physiological index is a dynamic process,mostly increasing first and then decreasing to the trend;different tissues of A.luridus respond to the stress treatment,and in most treatments.The leaf response was the fastest and the highest response value,and the root response was slower.4.This study reconstructed the metabolic pathway of hyoscyamine based on the transcriptome and metabolome,and found that the main alkaloid components of bellflower were mainly distributed in leaves and stems.5.Their change of transcription and metabolomes.of A.luridus that respond to drought,low temperature,high temperature,ultraviolet and other stresses.They can find up-regulated genes and up-regulated metabolites.The analysis found that the A.luridus responded to drought stress and the up-regulated genes were mainly carbohydrate metabolism genes such as sugar;the up-regulated substances were mainly amino acid substances.In response to low temperature stress,the A.luridus upregulate genes mainly related to phosphate signaling,chloroplast composition,carbohydrates and other carbohydrates;the main upregulation metabolites are lipids,sugar alcohols and phenolic acids.A.luridus cope with high temperature stress: the main up-regulated genes are heat shock protein,ribosome and other synthetic genes,and the main up-regulated metabolites are lipids and phenolic acids.The A.luridus should deal with ultraviolet treatment.The main up-regulation genes are related genes such as sugar and amino acid synthesis and transport,and the main up-regulation metabolites are lipids,flavonoids and phenolic acids.
