Recently,cancer has become a major threat to human health all around the world."Early detection,early treatment" and symptomatic treatment are the key to improve the cure and survival rate of cancer patients.Based on this,the combination of nanostructures with fluorescent probes on liquid biopsy markers could be developed to identify and monitor tumors in early stage.These nanoprobes could become powerful tools in disease diagnosis,prognostic prediction,and dynamic monitoring of therapeutic response-paving the way for personalized medical care.We proposed a dual-targeting magnetic-fluorescent nanobead(MB-MLP-EpCAM)which was composed of the antibody EpCAM and a proteinase(APN)-driven NIR fluorophore on Fe3O4 nanobeads to accurately isolate,enumerate and detect HCC-CTCs in one step.The dual-targeting nanobeads significantly improved the CTC-capture efficiency(85%)without scarificing cell viability(90%).Importanly,our nanoplatform were the first to achieve the realtime in vivo monitoring of CTCs in peripheral blood.We proposed a fluorometric "turn-on" Au nanoparticle(DHANs)with the conjunction of the tumor targeted moiety-alkynated dehydroascorbic acid(DHA)and a HIF 1?-switchable fluorometric aptamer.The introduction of tumor hypoxia microenvironment as a stimuli into the detection strategy could significantly improve the signal-to-background ratios and avoid the false-positive results.Besides,we designed the dual-functional "GSH-activable fluorescence" and "DHAtargeting" polymetric micelle(DPPP-nanoprobe).The nanoprobe was fabricated with a crosslinker L-lysine(PLys),a hydrophobic GSH-activable probe conjugated L-phenylalanine and a tumor targeting moiety,DHA-PEG-S-S.The nanoprobe was utilized as a safe,efficient,and precise means of CTCs identification.Besides,this nanoplatform could avoid the false negative signals due to the internal heterogeneity and be used extensively in both in vitro experiment and clinic online test.
