QUINO(1,2-C)QUINAZOLINES AND INDAZOLO(2,3-A)QUINOLINES: ANALOGS OF THE ANTITUMOR BENZO(C)PHENANTHRIDINE ALKALOIDS NITIDINE AND FAGARONINE

Abstract/Summary:
Several derivatives of the quino{1,2-c}quinazoline ring system have been prepared as analogs of the potent antitumor benzo{c}phenanthridine alkaloids nitidine and fagaronine, namely: 9,10-dialkoxy-1,2,3,4,12,13-hexahydro-1-oxoquino{1,2-c}quinazolinium perchlorates; 2,3,9,10-tetraalkoxyquino{1,2-c}quinazolinium perchlorates; 2,3,9,10-tetraalkoxy-12,13-dihydro-11bH-quino{1,2-c}quinazolines; 2,3,9,10-tetraalkoxy-12,13-dihydro-11bH-quino{1,2-c}quinazoline-6-(7H)thiones and 2,3,9,10-tetraalkoxy-12,13-dihydro-11bH-quino{1,2-c}quinazolin-6-(7H)ones. Two derivatives of the related indazolo{2,3-a}quinoline ring system were also synthesized, namely: 2,3-dimethoxy{1,3}dioxolo{4,5-g}indazolo{2,3-a}quinoline and 2,3,8,9-tetramethoxyindazolo{2,3-a}quinoline. The structures of these compounds were proven on the basis of spectroscopic data (ir, nmr, ms) and elemental analysis.
