Seven unique amino acid substitutions have been introduced into the first conserved region of the small subunit of ribulose bisphosphate carboxylase/oxygenase from Anacystis nidulans. Two substitutions in the small subunit ;Part II. A fast and efficient method for purification of DNA from various sources is described. The method utilizes efficient adsorbtion of DNA to silica particles in the presence of a chaotropic agent. The DNA purification is facilitated by the use of a combination of minicolumns, prepared from common filtered pipet tips, and use of a vacuum source, significantly facilitating the processing of numerous samples. Protocols are described for small and large scale plasmid preparations, phage DNA purification, gel elutions, and purification of DNA from solutions.
