Gene expression in the ruminant placentome

Abstract/Summary:
Little is known about the gene expression in the ruminant placentome throughout gestation. This research identified a number of novel genes as well as several previously identified genes that are expressed in the bovine placentome at day 90 of gestation in the cow.;A cDNA library was constructed by subtracting intercaruncular mRNA from caruncular cDNA using the Invitrogen Subtractor Kit (Invitrogen, San Diego, CA). This technique was used to enrich the cDNA library with sequences expressed by the caruncular tissue at day 90 of gestation. One-hundred and thirty-six cDNA clones were partially sequenced from the 3;Twelve genes characterized were classified as "housekeeping" genes due to their expression in all tissues examined. Clone ;Two genes were expressed specifically in the cotyledon and caruncle at day 90 of pregnancy, placental lactogen (clone ;The remaining ten genes were classified as cDNAs showing differential expression among uterine tissues and/or non-reproductive tissues. 88% of the 25 cDNAs isolated were not previously identified in the cow. These cDNAs will be useful as aids in further characterization of the bovine placentome and several may be useful as markers to study the bovine genome.
