Abstract/Summary:
Described herein is the discovery and characterization of an antibody biocatalyst for a 1,3-dipolar cycloaddition reaction between a nitrile oxide and various acrylamide substrates. Complete bimolecular reaction kinetics were carried out with antibody catalyst 29G12, substrate specificity studies, and thermodynamic parameters determined for the reaction. Structural studies were undertaken to explore the origin of catalysis and to suggest possible directions for improved catalytic proficiency.
