Objective: Observing the influence of the structure and function of autophagy body from skeletal muscle cells after a heavy loadtraining,analysis the changes of autophagy and the associated protein;As the breakthrough point,using Omi/Htr A2 to explore the molecular mechanism of skeletal muscle autophagy induced by heavy load training;To observe the effects of acupuncture which about skeletal muscle autophagy after heavy load training,and clarify the mechanism.Methods: The first study,the rats were divided into control group(C)and exercise group(E);the second study,the rats were divided into control group(C),placebo(D),inhibitor group(U),exercise group(E)and inhibitor exercise group(EU);the third study,the rats were divided into control group(C),acupuncture group(A),exercise group(E)and acupuncture exercise group(EA).E is a downhill running of heavy load,U is used inhibitor,A is intervention with acupuncture.Each group choose the time 0 h,12 h,24 h,48 h and 72 h after the intervention,using transmission electron microscope to observe ultrastructural changes of autophagy in the cells of the soleus.Using Western Blot method to detect protein expression of Omi/Htr A2,Hax-1,Beclin1 and LC3 in soleus;Using immunofluorescence technology fix position of LC3;With immune co-precipitation technique levels Hax-1 and Beclin1 protein binding.Results:(1)Body shape and quantity of autophagy are obvious change after a heavy load training in soleus muscle cells,and autophagy fluorescence enhanced about LC3,the expression of Beclin1 and LC3 increased transiently;(2)the expression of Omi/Htr A2 increased after a heavy load training,and the combine of Hax-1 and Beclin1 reduced.The expression of Omi/Htr A2 decrease after the intervention in ucf-101,Hax-1 and Beclin1 protein increased;(3)EU and EA can obviously decrease the phage in the soleus muscle cells induced by heavy load movement,cut the expression of Beclin1 and LC3,reduce the fluorescence intensity of LC3.Conclusions:(1)A large load eccentric training can induce autophagy of skeletal muscle cells,which enhancement can be achieved by Omi/Htr A2-Hax-1-Beclin1 way;(2)The ucf-101 can reduce the level of autophagy by inhibiting the cutting effect of Omi/Htr A2 to Hax-1,increasing the inhibitory effect of Beclin1;(3)Acupuncture by Omi/Htr A2-Hax-1-Beclin1 way to reduce the autophagy which induced by a large centrifugal load movement.
