Aim: Pterygium is an aggressive hyperplasia with excessive proliferation of corneal epithelial cells,which eventually invades the cornea.The main purpose of this study is to isolate stem cells from pterygium and validate the differentiation strategy of pterygium stem cells in vitro.In this dissertation,recent literatures and related studies in this field are briefly introduced,which prove that differentiation therapy plays a key role in the treatment of various clinical diseases.Currently,surgery is the principle treatment for pterygium.The discovery and application of differentiation therapy strategy can greatly reduce the surgical risk of patients,slow down the progress of pterygium and avoid the recurrence of pterygium.Methods: Pterygium stem cells were obtained by primary culture in vitro.Immunofluorescence staining was used to study stem cell markers and multilineage differentiation potential of pterygium stem cells.Results: Pterygium stem cells could be cultured and expanded steadily in vitro.They express a variety of molecular markers of stem cells and can be induced to differentiate into neurons,osteocytes and adipocytes in vitro.After being induced by reagents pterygium stem cell spheres express a variety of stem cells and epithelial-mesenchymal transformation markers.After differentiation induction,the proliferation and viability of pterygium stem cells were inhibited,suggesting that differentiation induction therapy strategy is effective and has broad application prospects in pterygium treatment.Conclusion: Pterygium-derived cells express a variety of stem cell markers and have the potential of multidirectional differentiation.After treated with induction medium in vitro,the proliferation of pterygium stem cells decreased,and differentiation induction could effectively inhibit the viability of pterygium stem cells,suggesting that differentiation induction therapy strategy is effective and has broad application prospects in pterygium treatment.
