Long non-coding BANCR has been reported to be tightly associated with tumorigenesis and development in various types of cancers. However, the expression pattern, biological functions and modulatory mechanisms of BANCR in pancreatic cancer(PC)remained unclear. In the present work, the carcinogenic activity and underlying mechanisms of BANCR on PC were explored in vitro. The results showed that BANCR was up-regulated in PC tissues and cell lines, and BANCR was related to tumor lymph node metastasis and stage.Besides,BANCR reinforced proliferation, invasion, and migration of both PANC-1 and SW1990 cells.Moreover,miR-195-5p was down-regulated in both PC tissues and cell lines.Based on the results of dual-luciferase reporter gene assay and q RT-PCR,miR-195-5p was a direct target of BANCR.Furthermore,miR-195-5p inhibitor reversed the effects of si-BANCR on proliferation and invasion of PANC-1 and SW1990 cells in vitro. In addition,BANCR played a vital role in activating the Wnt/?-catenin pathway by sponging miR-195-5p.Collectively,our study revealed that BANCR may play the role of oncogene in pancreatic cancer, which can promote the proliferation, invasion and metastasis of pancreatic cancer. This study elucidates the molecular mechanism that BANCR can regulate Wnt/ ?-catenin signaling pathway to promote the progression and metastasis of pancreatic cancer through competitive binding of miR-195-5p,which provides new ideas and potential targets for diagnosis and treatment of pancreatic cancer.
