OBJECTIVE:On the basis of clinical efficacy,this project,through theoretical research,cell experiment and animal experiment,discusses the intervention effect of Panax Notoginseng on lung cancer and its possible mechanism of anti-tumor,so as to provide theoretical basis for clinical use of Chinese herbal medicine Panax notoginseng in the treatment of lung cancer.METHODS:1.Theoretical research:through consulting a large number of references,we can understand the pathogenesis of lung cancer,the medicinal mechanism of Panax notoginseng,the relationship between blood stasis and the pathogenesis of lung cancer,and the research progress of tumor treatment genetics.2.Clinical research(retrospective analysis):Based on the retrospective analysis of clinical data,it is concluded that Panax notoginseng may play a role in the treatment of lung cancer;Combined with the statistical results of clinical data collection,further research and certification are carried out at the cellular level and animal level.3.Cell experiment:1)screening the best target gene CTSB interference group;2)The effect of Sanqi powder on lung adenocarcinoma cell line and the mechanism of its effect on cathepsin B were studied from the cell level.4.Animal experiment:A549 lung adenocarcinoma cells were used for subcutaneous tumorigenesis,which was divided into 5 groups:normal control group,tumor bearing model group,low dose intervention group,medium dose intervention group and high dose intervention group,compared with normal lung tissue and tumor lung tissue,the regulatory effect of Sanqi powder on the expression of CTSB gene and the mechanism of inhibiting the growth of lung cancer were studied at the animal level.Results:1.Theoretical research:After consulting a large number of references,the results are analyzed as follows:1)TCM believes that there is a certain correlation between blood stasis and lung cancer;2)There are some controversies in the treatment of tumor by Huoxue Huayu medicine:"fighting for the cause of blood stasis to treat tumor" and "promoting the spread of tumor by Huoxue Huayu".3)The study of tumor treatment genomics found that cathepsin B was highly expressed in tumor tissue,which may be related to the pathogenesis of tumor,and its corresponding gene was determined to be CTSB.2.Clinical research(retrospective analysis):Two groups of lung cancer patients were observed.It was found that the level of tumor markers(CEA,NSE and CYFRA21)and D-dimer and fibrinogen could be reduced in varying degrees after treatment with Sanqi powder.Among them,CYFRA21,D-dimer and fibrinogen improved significantly,but CT results showed no progress.The tumor in patients was stable and the quality of life was improved.3.Cell experiments:1)The expression of cathepsin B in A549 cells was inhibited by siRNA targeting cathepsin B after transfection;The results showed that the proliferation ability of A549 cells decreased and apoptosis ability increased,and the change of proliferation ability and apoptosis ability was dose-dependent.The expression of CTSB mRNA in A549 cells was down regulated by FQ-PCR,especially in high dose group;4.Animal experiments:l)Compared with the normal control group,the expression of cathepsin B mRNA and protein was high;2)Compared with the tumor model group,the growth rate of tumor in the intervention group was significantly slower and dose.dependent,and there was significant difference in tumor volume between the different groups(P<0.05);After the intervention of Panax notoginseng,the expression level of CTSB mRNA and CTSB protein in different groups of tumors decreased in varying degrees,which was dose-dependent;After intervention,Panax notoginseng can affect the level of tumor markers(CEA,NSE and CYFRA21)and D-dimer in serun of mice,reduce the content of D-dimer in serum of tumor bearing mice,and prolong the total survival period of mice in different degrees,especially in high dose group.CONCLUSION:Through theoretical research,clinical retrospective analysis and in vitro and in vivo experiments,it is confirmed that Panax notoginseng can inhibit the growth of lung cancer by acting on the CTSB gene,down regulating the expression of cathepsin B(CTSB)mRNA and protein,reducing D-dimer,changing blood viscosity and so on,so as to achieve the anti-tumor effect which provides a certain experimental basis for the clinical application of Panax notoginseng(powder)in the treatment of lung cancer.
