Study On The Pregnane Alkaloids And Their Anti-metastatic Activities Of Breast Cancer From Pachysandra Terminalis

Pachysandra terminalis Sieb.Et Zucc.is distributed in the southwestern region of China and Japan.It has been used as a traditional medicine against pain and stomach problems with pregnane alkaloids as its main bioactive chemical constitutes.Previous pharmacological research has revealed that these compounds produced a wide range of pharmacological effects including antioxidant,antiulcer,cytotoxic,anti-cancer,anti-metastasis activities,anticholinesterase as well as estrogen biosynthesis-promoting effects..As part of our ongoing research to identify bioactive steroidal alkaloids with anti-metastatic effects,nine novel(1–9)and forteen known(10–23)pregnane alkaloids were isolated from the whole herb of Pachysandra terminalis.We described herein the isolation,structure elucidation,and anti-metastatic effects of these compounds.In addition,their structure activity relationship was also discussed.Nine pregnane alkaloids(1-9)were isolated from the whole herb of Pachysandra terminalis,together with forteen known alkaloids(10-23).Their structures were elucidated on the basis of spectroscopic analyses.In addition,the compounds were tested for their ability to inhibit the migration of MDA-MB-231 cells induced by the chemokine epidermal growth factor(EGF).Compounds 1,5,7,9,12,and 17 presented significant anti-metastatic activities compared with the positive control,LY294002.This paper described the isolation,identification,and effects of compounds1-9 on the invasion of MDA-MB-231 breast cancer cells in vitro.Terminami-ne K [20α-dimethylamino-16β-hydroxy-3β-(3′α-isopropyl)-lactam-11α-isovaleryl-5α-pregn-1β,4β-diyl acetate(1)],Terminamine L [20α-dimethylamino-3β-(3’-isopr opylidene)lactam-5α-pregn-4-one(2)],Terminamine M [3β-dimethylamino-20α-s enecioylamino-5α-pregn(3)],Terminamine N [3β-dimethylamino-20α-senecioyla mino-5α-pregn-5-en(4)],Terminamine O [3β-methylamino-20α-senecioylamino-5α-pregn-5-en(5)],Terminamine P [3α-methylamino-20α-senecioylamino-5α-preg n-5-en(6)],Terminamine Q [20α-dimethylamino-16β-hydroxy-3β-senecioylamino-preg-5-ene(7)],Terminamine R [3β-benzoylamino-4α-hydroxy-5α-pregn(8)],Terminamine S [(Z)-3β-(dimethylamino)-17(20)-pregnene-5-en-16-one(9)]。...
