As traditional Chinese medicine,Acanthopanax senticosus is the dried root and rhizome or stem from the plant Acanthopanax senticosus(Rupr.Et Maxim.)Harms of Acanthopanax genus in Araliaceae family.In recent years,the chemical constituents and pharmacological effects of Acanthopanax senticosus have been mainly focused on saponins and flavonoids,and they have been widely used in the clinical treatment for cardiovascular and cerebrovascular diseases.Polysaccharide from Acanthopanax senticosus is one of the important active ingredients for Acanthopanax senticosus.As the purification process of polysaccharide components is very complicated,the purification process is complex and the purity is not ideal,the confirmation of polysaccharide from Acanthopanax senticosus in terms of composition,structure-activity relationship and action mechanism is limited.In this study,the chemical constituents,the anti-inflammatory effect,the analgesic effect,the protective effect of drug-induced liver injury,the adjuvant therapeutic effect of tuberculous pleurisy,the anti-tumor effect,the anti-aging effect of,and the role in improving learning ability and memory of polysaccharide from Acanthopanax senticosus in Changbai Mountain(ASPS)were studied intensively.At the same time,the preparation technology of ASPS capsule was discussed,in order to provide favorable reference for the clinical application of ASPS.The results are as follows:1.In this study,through the extraction and separation of ASPS,the chemical constituents were analyzed by high performance liquid chromatography and gas chromatography.The results showed that,after extraction and separation analysis,the yield of acanthopanaxe E was 93.1%,the yield of ASPS was 96.7% and the relative molecular weight was 10 KD.The evenly,symmetrical,single peak of the analysed ASPS sample revealed its homogeneous composition.The monosaccharide composition of ASPS was demonstrated to be(Arabinose: Xylose: Glucose: Mannose)= 7.1:22.3:7.6:1.0.2.With xylene-induced auricle swelling in mice,carrageenan-induced toe swelling in mice,increased peritoneal capillary permeability in mice induced by acetic acid,and cotton ball induced granuloma in mice as models,the anti-inflammatory effect of ASPS was studied.The results showed that ASPS effectively inhibited the xylene-induced auricle swelling in mice,carrageenan-induced toe swelling in mice,increased peritoneal capillary permeability in mice induced by acetic acid,and cotton ball induced granuloma in mice.At the same time,ASPS effectively improved the immune organ status of mice and regulated the levels of immune cytokines in different anti-inflammatory models.3.By thermal stimulation of the impact of pain threshold and acetic acid induced writhing reaction,the analgesic effect of ASPS was studied.The results showed that ASPS increased the pain threshold and analgesic rate of heat-stimulated mice,reduced the number of writhing induced by acetic acid and increased the inhibition rate of writhing response.At the same time,ASPS significantly improved the condition of immune organs and regulate the level of cytokines in different models of analgesia.4.With experimental tuberculosis mice as models,the role and the mechanism of ASPS for antituberculosis drug-induced liver injury were studied.The results showed that ASPS had better liver protection effect on experimental tuberculosis mice with antituberculosis treatment.The mechanism may be through the regulation of serum levels of cellular immune factors and T lymphocyte subsets of spleen cells immune levels of the experimental tuberculosis mice,so as to improve their immune capacity and then to achieve its role in protecting liver.5.With tuberculous pleurisy rats as models,the treatment effect and its mechanism of ASPS were studied.The results showed that ASPS reduced tuberculosis pleurisy pleural effusion and pleural thickness,and improved white blood cell count and biochemical indicators of pleural effusion of the tuberculous pleurisy rats.In addition,ASPS also improved and regulated the cellular immune factors in tuberculous pleurisy rats,which may be one of its mechanisms of action for the treatment of tuberculous pleurisy.6.With S180 sarcoma,H22 hepatoma and U14 cervical cancer solid tumor and ascites tumor mice as models,ASPS antitumour effect and its mechanism were studied,and its toxic effects were analyzed.The results showed that ASPS had good inhibitory effect on the solid tumor of S180 sarcoma,H22 hepatoma and U14 cervical cancer tumour-bearing mice,and the inhibition rates of tumour-bearing mice were increaded significantly.Meanwhile,ASPS increased the immune organ index,regulated the immune cytokines of the tumour-bearing mice,so as to improve the immunity of the body and enhance their resistance to disease.ASPS extended the survival time of S180 sarcoma,H22 hepatoma and U14 cervical cancer ascites tumor mice,and improved their life extension rates actively.Acute toxicity test results show that ASPS was an actually non-toxic substance.7.With D-galactose induced aging mice as models,ASPS anti-aging effect and its mechanism were studied.The results showed that ASPS effectively improved the body weight and liver index of aging mice induced by D-galactose,and improved the body condition.ASPS also effectively improved the activities of superoxide dismutase(SOD),glutathione peroxidase(GSH-PX)and catalase(CAT)in the serum,brain and liver,while decreasing malondialdehyde(MDA)content in brain and liver tissue of model mice.It is further confirmed that ASPS plays an anti-aging role in enhancing the antioxidant enzyme activity of the body and reducing the cell damage caused by lipid peroxidation.8.With scopolamine induced acute memory impairment rats as models,by dark-avoidance test and Morris water maze test,ASPS enhancing memory effect and its mechanism were studied.The results showed that in the dark-avoidance test,ASPS effectively prolonged the avoidance latency and light box latency,and reduced the number of dark avoidance errors of memory impairment rats.In the Morris water maze test,ASPS prolonged the avoidance latency and the original platform retention time,and increased the number of platform exploration of memory impairment rats.At the same time,ASPS increased the activity of superoxide dismutase(SOD)and decresed the content of malondialdehyde(MDA)in the brain tissue and serum of model rats.ASPS improved the activities of acetylcholinesterase(AChE)and acetylcholinesterase(ChAT)of the central cholinergic system,so as to enhance learning ability and memory.9.By the screening of excipients,the screening of granulation process conditions,the study on drug powder formability,the preparation of capsule and the detection of capsule preparation,the preparation of ASPS capsule was studied.The results showed that corn flour was more suitable as ASPS auxiliary material for capsule dosage form preparation.ASPS particle shape and granulation effect showed good state with 90% ethanol concentration,1:1 ratio of solid to liquid,60? drying temperature,1h drying time.In the production and storage process of ASPS capsule,the relative humidity should be strictly controlled at 62% or below in order to ensure the quality of the powder.For ASPS capsule,the moisture content was 7.04%,the average content was 0.4315 g,the average disintegration time was 17.43 min,which all meet the relevant provisions in Chinese Pharmacopoeia(2015 version).
