α-Enoase(ENO1)gene is a glycolytic enzyme that catalyzes the conversion of2-phosphoglycerate to phosphoenolpyruvate.ENO1 is widely expressed in various human tissues and plays an important role in the development and progression of various tumors.However,the role of ENO1 in gastric cancer is unclear.Thus,the specific function and molecular mechanism of ENO1 in gastric cancer need to be further studied.ObjectiveThe aim of present study are to detect the expression level of ENO1 in human gastric cancer tissues and matched non-cancer tissues,explore its biological function in gastric cancer,and elucidate the related mechanism.Method1.Immunohistochemical staning was used to detect the expression of ENO1 in gastric cancer tissues and adjacent tissues and the results were compared with those in the TCGA database.Kaplan-meier method was used to analyze the correlation between ENO1 expression and prognosis of gastric cancer patients.2.RNA interference technology was applied to knockdown the ENO1 expression,and the effect of ENO1 knockdown on the biological function of gastric cancer cells was detected by CCK8,clone formation,apoptosis experiment,cell cycle experiment and scratch experiment.3.Genome-wide mRNA expression abundance in gastric cancer cells MGC-803 was detected by Affymetrix human whole gene expression spectrum microarray with or without ENO1 konckdown.Application of Ingenuity pathway analysis was to analyze differentially expressed genes and help us to find the downstream potential candidate genes.Real-time PCR was used to detect the changes in mRNA expression level if the downstream pathway genes after ENO1 knockdown.Results1.The expression of ENO1 was significantly elevated in gastric cancer tissues than that in adjacent tissues,and its expression level in gastric cancer cells was higher than that in normal gastric epithelial cells.The survival rate of gastric cancer patients with high expression of ENO1 was lower than those with low expression of ENO1.2.Knockdown of ENO1 could markedly inhibit the proliferation of gastric cancer cells,induce the apoptosis of gastric cancer cells and lead to cell cycle arrest in S phases,which indicated that ENO1 may affect the proliferation of gastric cancer cells by inducing apoptosis.Furthermore,ENO1 silencing markerdly suppressed the migration ability of gastric cancer cells.3.Knockdown of ENO1 could lead to expression changes of 681 downstream genes by detection of mRNA expression profile chip.And by IPA analysis we found that ENO1 knockdown could inhibit the activity of HGF/PI3K/AKT/c-Fos signaling pathway.Moreover,ENO1 knockdown could increase the mRNA expression level of MAP3K20 and DET1 and reduce the mRNA expression level of c-Fos and SBDS.Conclusions1.ENO1 is involved in the development of gastric cancer,and ENO1 may act as an oncogene in gastric cancer.2.ENO1 may promote the proliferation and migration of gastric cancer cells through HGF/PI3K/AKT/c-Fos signaling pathway.3.ENO1 is expected to become a potential diagnostic marker of gastric cancer,and provide theoretical basis for deep understanding of the mechanism of gastric cancer and the screening of drug targets.
