Expression and clinical significance of Linc00152 in gallbladder cancer tissuesObjective To investigate the expression of Linc00152 and its significance in gallbladder cancer tissues.Methods Linc00152 expression was determined in 35 GBC and adjacent tissues by real-time quantitative PCR.The correlation between Linc00152 expression in GBC and clinicopathologic parameters was analyzed by using a Pearson's Chi-square test.The Kaplan-Meier test and Cox proportional hazards model were used to measure the survival in GBC patients.Results The expression of Linc00152 was significantly increased in GBC tissues.Linc00152 expression was significantly associated with histological.The median survival time in the high-expression subset shorter than the low-expression subset.Multivariate analyses by using the COX proportional hazard regression model further confirmed that Linc00152 overexpression was negatively correlated with overall survival.Conclusion Linc00152 expression was associated with histological grade.Linc00152 expression is an independent prognostic factor in GBC patients.Part ? The biological function and molecular mechanism of Linc00152 in gallbladder cancerObjective To study the biological function and molecular mechanism of Linc00152 in gallbladder cancer.Methods Cell proliferation ability and invasiveness were evaluated by using CCK8 assay,colony formation,transwell invasion and migration assay in vitro.Knock down the Linc00152 of NOZ cells by using CRISPR/Cas9 systems.Subcutaneous xenograft tumor models in nude mice by using Linc00152-knockdown and wild type NOZ cells.Western blot analysis were performed for the molecular mechanism of Linc00152 in gallbladder cancer.Results Linc00152 in NOZ and GBC-SD cells was knocked down by si RNA and CRISPR/Cas9 systems.After silencing Linc00152 in GBC cell lines,the proliferation,migration and invasion was significantly decreased both in vitro and in vivo.Furthermore,ERK/MAPK pathway was found to be inactivated in GBC cell lines after Linc00152 knockout.Conclusion Linc00152 enhanced cell proliferation,migration and invasion by activating ERK/MAPK pathway.
