Objectives: To study the expression and intracellular localization of protein ANLN,SHMT2,PHK? in colorectal cancers(CRC);analyze its correlation with clinicopathological features,as well as prognosis;Study the biological functional roles and underlying mechanism of protein SHMT2,PHK? in CRC cells lines.Methods: q PCR,western blot assays were performed to compare expressions of ANLN m RNA and protein in CRC tissues and matched normal mucosa;tissue microarrays and immunohistochemical staining were performed to detect expression and intracellular location of protein ANLN,SHMT2,PHK? analyze its correlation with clinicopathological characteristics and prognosis in CRC patients;Lentivirus infection to establish stable CRC cell lines expressing SHMT2 sh RNAsconstantly;proliferation,cell cycle and colony formation of each stable cell lines were detected.Results: ANLN,SHMT2,PHK? overexpression were found in CRC tissue compared to normal mucosa;higher expression of ANLN,SHMT2,PHK? was independent prognostic factor in patients with CRC;SHMT2 and PHK? knockdown impaired proliferation and colony formation of colorectal cancer cells.Conclusions: Our result revealed that high expression of ANLN,SHMT2,PHK? were independent prognostic factor in patients with CRC;SHMT2 regulates CRC cell proliferation,colony formation and cell cycle through UHRF1 gene;PHK? regulates CRC cell proliferation and cell cycle through p53-induced cell-cycle arrest.
