Study On The Autophagy Function Of Bronchial Asthma Rats With Syndrome Of Cold Retention Accumulation In Lung And The Intervention Mechanism Of Chinese Herbs Based On Theory Of Celestial Qi Communicating With Lung

Objective: To explore the theory of celestial qi communicating with lung and its theoretical origin and main contents,analyze the characteristics of haze and its relationship with syndrome of cold retention accumulation in lung,explore the disease mechanism of asthmatic rats with syndrome of cold retention accumulation in lung induced by cold stimulation in smoky environment,and the intervention mechanism of Chinese herbs on autophagy in lung tissue.Methods: In this paper,theoretical and experimental methods were used.(1)Theoretical study: By using methods of literature research and theoretical analysis,we explored the theory of celestial qi communicating with lung and its theoretical origin and main contents,relations between lung and environment,characteristics of haze and its relationship with syndrome of cold retention accumulation in lung.Meanwhile,we researched the relationship between lung yang and syndrome of cold retention accumulation in lung,the connotation,etiology,pathogenesis and treatment of the syndrome.(2)Experimental study: With egg albumin sensitization,cold stimulation,smoke stimulation and fatigue stimulation,we built model of bronchial asthma with syndrome of cold retention accumulation in lung in smoky environment.With the intervention of Xiaoqinglong decoction and Wenyang Huayin formula,we observed the relevant test indexes.Results: Combined with both traditional view of etiology and pathological model of modern medicine,we successfully replicated the model of bronchial asthma rats with syndrome of cold retention accumulation in lung in smoky environment.We made a comparative research about changes of inflammatory cytokines in lung function and serum in rats of different groups and found that haze can aggravate the incidence of bronchial asthma with syndrome of cold retention accumulation in lung,and confirmed the pathological mechanism of haze aggravating bronchial asthma.The results shows that smoking can increase the inhalation resistance(Ri)and exhalation resistance(Re)and decrease the compliance(Cdyn)of lung function in bronchial asthmatic rats,resulting in aggravated asthma symptoms in rats.The mechanism may be that smoking can induce the level of serum IL-13 and TGF-?1,reduce the level of IFN-?in asthmatic rats with syndrome of cold retention accumulation in lung,and increase the inflammation response of the airway.We observed the asthmatic rats with syndrome of cold retention accumulation in lung and intervened with Chinese herbs and examined such indexes as the lung function,pathological section and inflammation factor.The results showed that Wenyang Huayin formula can promote the level of IL-10,IL-18,and IFN-?,reduce degrees of IL-4,IL-5,IL-13,TGF-?,and TNF-?in the blood,thereby inhibiting allergic reactions and regulating the occurrence and development of inflammation.The mechanism maybe that Wenyang Huayin formula can inhibit the secretion of extracellular matrix and the collagen deposition process of pulmonary fibroblasts,slow airway remodeling and inhibit occurrence and development of asthma by reducing the expression of rat autophagy gene Atg and autophagic protein Beclin1 and LC3-a/b.Conclusion: Celestial qi refers to the clean air of nature,which is connected with lung qi.Haze is a compound pathogenic factor.It is characterized by dispersion,and often combines with dampness and heat pathogens to invade the lung and induce diseases.There are similarities in the nature and pathogenic characteristics of tobacco smoke and haze,and the main components that the particles contained in both of them are PM2.5.The normal transport of body fluid needs the transpiration of yang qi.If lung yang is sufficient,the lung perform the normal functions of dispersing and descending,and thus body fluid can transport properly.The pathological mechanism of bronchial asthma with syndrome of cold retention accumulation in lung is lung yang deficiency,which causes the accumulation of phlegm,the increase of serum levels of IL-13 and TGF-?1,and the decreased level of IFN-?,thereby aggravating the airway inflammation.The mechanism is that Wenyang Huayin formula reduces the level of airway inflammation,reverses airway remodeling,and regulates the high airway response by reducing the expression of the autophagy gene Atg and the levels of protein Beclin1 and LC3.
