Research purposes:To explore the effect of Qi-dan-di-huang Decoction on Diabetic Nephropathy rat model via NF-?B signaling pathway,and investigate its kidney protective effect and mechanism.Method:Part ?,establish Diabetic Nephropathy rat model induced by intraperitoneal injection of streptozotocin(STZ),and explore the effect of Qi-dan-di-huang Decoction on physiological and biochemical indexes of model rats.The body weight,blood glucose,24h urinary albumin,serum creatinine,blood urea nitrogen,24h U-vol,KW/BW values were measured.Part ?,study the effect of Qi-dan-di-huang Decoction on the kidney of rats with diabetic nephropathy.HE staining,Masson staining,and PAS staining were performed on paraffin sections to investigate the protective effect of Qi-dan-di-huang Decoction on the kidneys.Part ?,study the effect of Qi-dan-di-huang Decoction on NF-?B signaling pathway in diabetic nephropathy rats.The effect of Qi-dan-di-huang Decoction on the p-p65,p65,p50,p-?B?,I?B?,I?B?,I???/?,TGF-?,?-SMA and other proteins in the NF-?B signaling pathway was examined by Western blotting.Results:Part ?I:The T1DM rat model was successfully established,and Qi-dan-di-huang Decoction had obvious improvement on physiological and biochemical indicators of the model rats.Part ?:The results of kidney tissue HE staining,Masson staining and PAS staining showed that compared with normal rats,the diabetic nephropathy rats had severe glomerular fibrosis,increased extracellular matrix accumulation,mesangial thickening,capillary wall thickening,and a lot of glycogen protein deposition in the glomerular and tubular cells,visible nodules,unclear structure.The degree of glomerular fibrosis was improved in the fenofibrate group and the Qi-dan-di-huang Decoction group.Their glomerular mesangium was slightly thickened and their capillary wall was slightly thickened in the fenofibrate group and the Qi-dan-di-huang Decoction group.A small amount of glycogen was deposited in the glomerular and tubular cells,and the glomerular and tubular structures became normal in the Qi-dan-di-huang Decoction group.Part ?:The levels of IL-6,IL-1? and TNF-a in the serum of rats were detected by Elisa in each group.The_serum levels of IL-6,IL-1? and TNF-? in the rats of the fenofibrate group and the Qi-dan-di-huang Decoction group were significantly lower than the model group.The expression of IL-6 in serum of Qi-dan-di-huang Decoction group was significantly lower than that of fenofibrate group rats.At the same time,the expression of p-p65,p65,p50,p-?B?,I?B?,I?B?,I???/?,TGF-? and ?-SMA in NF-?B signaling pathway was detected by Western blotting.Compared with the normal group,the levels of p50,p65,I?B? and I?B? was not changed and the expression of I???/?,p-p65,p-I?B?,TGF-? and ?-SMA was significantly increased in the model group.Compared with the model group,the expression of I???/?,p-p65,p-I?B?,TGF-? and ?-SMA in the kidneys of the fenofibrate group and the Qi-dan-di-huang Decoction group was significantly lower than that of the model group.Conclusions:Drug intervention based on successful modeling,we found that Qi-dan-di-huang Decoction could significantly improve the physiology and biochemistry of model rats in terms of histology and other indicators.It could alleviaterenal fibrosis in rats with diabetic nephropathy,and reduce glycogen protein deposition in glomeruli and tubular cells.The study found that Qi-dan-di-huang Decoction significantly inhibited the production and release of inflammatory cytokines in diabetic nephropathy model rats.The levels of I???/?,p-p65,p-I?B?,TGF-? and ?-SMA in the diabetic nephropathy model group were significantly increased.However,the treatment with Qi-dan-di-huang Decoction could significantly reduce the expression of I??q/w,p-p65,p-I?B?,TGF-? and a-SMA in DN rats.It was demonstrated that Qi-dan-di-huang Decoction could significantly inhibit the activation of NF-?B signaling pathway in diabetic nephropathy model rats.Therefore,it can be concluded that Qi-dan-di-huang Decoction protects the kidney by inhibiting the activation of NF-?B signaling pathway,inhibiting the production and release of inflammatory cytokines,improving the physiological and biochemical characteristics of renal tissues and the renal histology.
