Association Between Clusterin Gene Polymorphisms And Epilepsy In Han Chinese Population

Objective:Clusterin plays an important role in the immune inflammatory mechanism of the nervous system,but the correlation between clusterin gene SNPs and epilepsy is still unclear.This study is the first time to explore the correlation between single nucleotide polymorphisms(SNPs)of clusterin gene and epilepsy and antiepileptic drugs.Methods:249 epileptic patients(case group)and 289 noramal subjects(control group)were included in a retrospective study.According to relevant literatures,three SNPs(rs11136000,rs9314349 and rs9331949)of the clusterin gene were selected.The differences of genotype frequency of the three loci were compared in the case group and the control group,and further stratified studies were conducted according to gender,age of onset,type of onset and drug resistance.According to the results of the retrospective study,152 epilepsy patients and 130 normal subjects were prospectively included.Patients with epilepsy were treated with valproate monotherapy,and were divided into effective group and drug resistant group according to the curative effect.First,the distribution differences of the genotype frequencies of the three SNPs in each group were compared.Then,the differences of valproate drug doses and blood drug concentrations in patients with epilepsy of different genotypes were compared by recording the dosage of sodium valproate and measuring the blood drug concentration of sodium valproate.Results:In the retrospective study,there was no statistical difference in the genotype frequency of the three SNPs of the clusterin gene between the case group and the control group(p>0.05).In the prospective study,there was no statistical difference in the distribution of genotype frequency of the three SNPs in each group(P>0.05).The blood concentration per kg and the dose per kg in the sodium valproate resistance group were higher than those in the effective group,and the difference was statistically significant(p<0.05),but there was no statistically significant difference in drug dose and blood concentration among patients with different genotypes(P>0.05).Conclusion:In this study,the association between three SNPs of the clusterin gene rs11136000,rs9314349 and rs9331949 and epilepsy was not found,and the gene polymorphism was not related to the drug resistance of sodium valproate.The relationship between single nucleotide polymorphisms and epilepsy needs to be further explored.
