This research is based on the cancerous toxin pathogenesis theory of Prof.Zhou Zhongying.We taked Xiao'ai Jiedu Prescription combined with radiotherapy to treat the patients with Esophageal cancer(EC),at the same time,the serum miRNAs were checked and analysed.The theoretical basis and the research results were summarized.Research objectives:The standardization of clinical concurrent controlled trials were conducted to observe the effect of Xiao'ai Jiedu Prescription combined with radiotherapy in treating Esophageal cancer patients(no surgery,II and III stage).Meanwhile,the traditional clinical efficacy indicaters(such as the tumor size,the level of tumor markers,physical condition,clinical symptoms,karnofsky score and the quality of life)were assessed,the clinical efficiency and the toxic side effects were also evaluated.In order to judge that whether Xiao'ai Jiedu Prescription combined with radiotherapy can ameliorate the quality of life and improve the clinical efficacy and reduce side reactions in patients.The experimental study was mainly to check the serum miRNAs of Esophageal cancer cases and healthy individuals,compare and analyze the differences.The aim is to investigate serum miRNAs profile for early Esophageal cancer diagnosis,then contrast the variations before and after treatment and discuss the michanisms of anti-tumor of Xiao'ai Jiedu Prescription,then provide theoretical foundation for clinical application.The ultimate objective is to improve the therapeutic effect of Esophageal cancer.Research methods:In clinical observation,60 patients who met criteria were divided into treatment group and control group,30 in the treatment group(those who received Xiao'ai Jiedu Prescription combined with radiotherapy),30 in the control group(those who received radiotherapy only).The efficacy were evaluated after treatment.At the same time,the karnofsky score,clinical symptoms,the tumor size,the level of tumor markers and side effects were observed in two groups.Using qRT-PCR to perfom expression of peripheral serum miRNAs between Esophageal cancer patients and healthy person.Identified miRNAs were further validated using qRT-PCR.The miRNAs were checked again after treatment,in order to provide some comparison analysis of the miRNAs before and after treatment.Spass 20.0 was employed to deal with these databases.Research results:The treatment group displayed more effective in reducing tumor markers,improving physical condition and relieving clinical symptoms(P<0.05).There was no obivious difference between the two groups in the objective curative effect and the side effects(P>0.05).Consequently in experimental study,a total of 16 miRNAs were identified,including 9 upregulated miRNAs and 7 downregulated miRNAs.To confirm the microarray findings,five serum miRNAs were assessed by qRT-PCR.The results showed that final tendencies were unanimous with microarray outcomes.Therefore,we focused on miR-223-3p and miR-151a-5p which were the miRNAs with consistently significant upregulation.The receiver operating characteristic(ROC)curve analysis indicated that miR-223-3p and miR-151a-5p could be used as biomarkers for Esophageal cancer diagnosis.The presentation standards of these two miRNAs in serum of Esophageal cancer patients were connected with the lymphatic metastasis and the tumor depth.The expression of miR-223-3p and miR-151a-5p reduced significantly after treatment,the miRNA level of treatment group was lower than that of control group.Research conclusions:Xiao'ai Jiedu Prescription combined with radiotherapy can improve physical condition and the quality of life,relieve the clinical symptoms,play an effect of reducing toxicity and dose not increase the side effects of radiotherapy.It is worthy of clinical use.The miR-223-3p and miR-151a-5p are upregulated in human Esophageal cancer.Xiao'ai Jiedu Prescription combined with radiotherapy can downregulate the expression level of these two miRNAs.
