Objective: To breed,reproduce and identify of the mice which is knockouted the H2-Eb1+H2-Ab1,homozygous for double H2-Eb1(-/-)+H2-Ab1 double wild type(-/-)and H2-Eb1(+/+)+H2-Ab1(+/+)mice of double knockout mice(H2-Eb1+H2-Ab1)for sensitization and excitation,by building a double gene(H2-Eb1+H2-Ab1)knockout mice model of allergic rhinitis disease research H2 Eb1,H2 Ab1 susceptibility to allergic rhinitis.Methods: the mice were bred and bred by the gene knockout mice and the mice were bred and bred.The DNA of rat tail was extracted and the genotype was identified by PCR.Hybridized mice were used to reproduce and protect species,and the wild type and homozygous mice were randomly divided into groups,models and lung tissues for genotype identification.Observe different groups of mice after building behavior score,HE staining of nasal mucosa and thymus tissue toluidine blue stain,serum Specific IgE,IL-2,IL-13,IL-17 A antibody levels,IL-2,IL-13,IL-17 A antigen expression level in the nasal mucosa.Results: the mice were successful in breeding,with two homozygous H2-eb1(-/-)+H2-ab1(-/-),20(20%),dual-wild-type H2-eb1(+/+)+H2-ab1(+/+)26(26%),and no genotype of 54(54%).H2-eb1+H2-ab1 double genes were successfully obtained for knockout homozygous mice,and the breeding,breeding and identification methods were established,and H2-ab1 and H2-eb of mice in the experimental group(KO-OVA)were knocked out.KO-OVA group compared with control group(WT-VOA)group,nasal mucosal tissue pathological changes is light,the behavior of mice,eosinophils in the nasal mucosa and thymus tissue mast cell count,serum OVA-Specific IgE,IL-13,IL-17 A in the nasal mucous membrane antigen expression degree is reduced to some extent,IL-2 antibody levels in serum,degree of IL-2 antigen expression in nasal mucosa increased to different extent.Conclusions: using female and male h2-eb1+ h2-ab1 double gene knockout hybrid mice.Not only H2-eb1+H2-ab1 double gene knockout homozygous mice,but also for long-term breeding and reproduction;The establishment of the ko-ova and wt-voa model of allergic rhinitis was successful,and Th1/Th2 cytokine imbalance was present in the two groups of mice.The susceptibility of the ko-ova group to allergic rhinitis decreased.
