BackgroundPolypoidal choroidal vasculopathy is one of the most important differential diagnosis of age-related macular degeneration,whose pathogenesis still remains unclear.Recent researches has revealed the importance of inflammation in wAMD and that improper regulation of the complement system might be related to its pathogenesis.CD11b+ monocytes and CD200+ monocytes were involved in the progress of wAMD via regulating the secretion of VEGF and the activity of microglia.Some complement proteins are proven expressing differently in wAMD patients,including CD21?CD35?CD46?CD55?CD59.However,most of these researches mentioned were not aiming at PCV.PurposeTo investigate surface expression of CD11b and CD200 on circulating monocytes in patients with PCV,wAMD and healthy components.To investigate surface expression of CD21?CD35?CD46?CD55?CD59 on circulating lymphocyte,monocytes and granulocyte.Briefly understand the relationship between PCV and inflammation environment and complement system.MethodThis is a case-control study.Recruit patients with PCV and wAMD,as well as age-matched healthy people.Make sure participants are not undergoing a current inflammatory reaction.Fresh circulating blood was stained with fluorescent antibodies for flow cytometric analyses.We compared the percentages of CDllb+,CD200+ and CDllb+CD200+ monocytes between groups of diagnosis.We also compared the percentages of CD21+.CD35+,CD46+.CD55+.CD59+lymphocyte,monocytes and granulocyte between groups of diagnosis.ResultCD200+ monocytes were decreased both in patients with PCV and in patients with wAMD.CD11b+ monocytes were increased both in PCV and in patients with wAMD.There was no significant difference between PCV and w patients concerning CDllb+CD200+ monocytes.There were several differences between CD21+.CD35+,CD46+.CD55+.CD59+ lymphocyte,monocytes and granulocyte between groups of diagnosis.ConclusionsCD11b+ and CD200+ monocytes were connected with PCV pathogenesis,via regulating the secreting of VEGF and the formation of CNV.PCV and wAMD might be both results of aging and improper regulation of immune functions.The complement system is palying an important role in the pathogenesis of PCV and wAMD.
