Objective:To explore the expression of microRNA-493-5p and synaptic vesicles of target genes associated membrane protein 2(VAMP2)regulation,control of cell proliferation,metastasis of liver cancer,promote apoptosis.Micro RNA-493-5p for the future as early diagnosis of liver cancer and provide a new treatment.Methods:Build HepG2 cancer cell lines and normal liver cells as a control group,the use of molecular biotechnology will pc DNA3.1-VAMP2 and miRNA-493-5p mimic transfection Hep G2 liver cancer cell lines,then the miR-493-5p in liver cancer cells and VAMP2 difference expression of health statistics analysis;In hepatocellular carcinoma cell line Hep G2 transfection in miR-493-5p simulation(mimic)to raise miR-493-5p expression,the impact on the cancer cells to a series of biological functions;Through common transfection miR-493-5p simulation(mimic)and pc DNA3.1-VAMP2,they work together on the impact of cancer cells.Using bioinformatics forecast online website Target Scan forecast miR-493-5p,VAMP2 may miR-493-5p is the main target genes.Results: MiR-493-5p expressed in liver cancer cells.Then we are in a series of molecular biology experiments,such as determined by MTT,flow cytometry technique and Transwell experiment,these get sexual experimental results indicate that overexpression of miR-493-5p expression makes liver cancer cells was significantly inhibited the proliferation and migration of ability.Successful build miR-493-5p mimic the transfection of tumor cell lines,pc DNA3.1 VAMP2 transfection of tumor cell line model.High expression of miR-493-5p directly inhibit the growth of tumor cell lines and proliferation.Conclusions: In hepatoma cell HepG2 cell lines,the miRNA-493-5p gene can be expressed through regulating VAMP2 to suppress the growth of tumor cells and induce the apoptosis of ageing.
