Association Between 18F-FDG PET/CT SUVmax And The Expression Of SET8 Protein In Patients With Laryngeal Cancer Of Different Survival Stages

Objective:This study based on the preoperative patients with laryngeal carcinoma PET/CT image data analysis,read the SUVmax value,and the postoperative histopathological SET8 and wild-type p53 protein expression,the relationship between the SUVmax,SET8 and wild-type p53 protein expression and the influence of survival.The effect of SET8 gene on proliferation,apoptosis and invasion and metastasis of laryngeal cancer cells was discussed by establishing human laryngeal cancer cell model.Methods:The expression of SET8 and wild-type p53 protein in 25 cases of laryngeal carcinoma were examined by immunohistochemistry.The relationship between SET8 expression,SUVmax and prognosis of patients with laryngeal carcinoma were analyzed.Establish a stable human laryngeal carcinoma cell line(TU212)and build sh-SETD8-TU212 cell lines through lentivirus infection.The wound scrape assay were used to detect the ability of TU212 cells migration.Brd U assay was used to detect the proliferation ability of TU212 cells.Flow cytometry method was used for apoptosis assay.RNA-seq and Western blot methods was for the expression of p53 in sh SETD8 cells.Results:25 people with laryngeal cancer surgery and without chemotherapy,radiotherapy and targeted therapy,did FDG PET/CT examination beforesurgery,postoperative pathological types of all cases were squamous cell carcinoma.The median survival time was 25 months and the overall survival rate was 56%.SUVmax,SET8 and p53 were all unrelated to the gender,age,primary site and treatment mode of the patient,and were correlated with clinical staging and pathological grading(P < 0.05).The overall survival rate was not correlated with the patient's gender,age,primary site and treatment.The expression of SETD8 was significantly increased in laryngeal carcinoma tissues,and the difference was statistically significant(P <0.01).With the increase of histological grade,the degree of tumor differentiation was lower and the expression of SETD8 was higher.By calculating the relative expression of wild-type p53 protein in laryngeal cancer cells,the total expression of wild-type p53 protein in laryngeal cancer cells was significantly reduced(P < 0.01).The mean SUVmax was11.6.When the mean SUVmax was defined as the cutoff,there was a significant difference in mean survival between the higher(?11.6)and lower(<11.6)SUVmax subgroups.?n the survival study:The survival rate of early laryngeal carcinoma(clinical ?+?)was 84.6%,while the survival rate of local advanced laryngeal carcinoma(stage ?+?)was only 25%.The difference is significant(p < 0.01);and the survival rate of high differentiated laryngeal carcinoma was 87.5%,and the survival rate of the pathologicallow-differentiated laryngeal cancer was 41.2%(p < 0.05).SUVmax < 11.6survival rate was 75.0% compared with SUVmax's survival rate of 11.6and 22.2%(p < 0.05).?t was divided into 2 groups according to the SET8 expression,low expression(negative,+),and high expression(+ +,+ + +),the low expression group survival rate was 75%,and high expression group survival rate was 22.2%,the statistical difference,(P < 0.05);its low expression of p53 group survival rate was 33.4%,and high expression group survival rate was 90%,the statistical difference(P < 0.05).The results of Western blot test showed that the expression level of SETD8 in sh SETD8 cells decreased significantly(P < 0.01)compared with that in the negative control group.The proliferation ability of TU212 cells decreased significantly,and the percentage of S stage cells decreased from 1.81 to 0.77 by knocking down SET8 gene(P<0.01).The migration distance of the laryngeal cancer cells after the down-regulated SET8 gene was 6h 0.015 mm vs.0.25 mm compared with the control group.12h0.05 mm vs.0.485 mm,24h 0.085 mm vs.0.63 mm significantly reduced(P< 0.01).Cell apoptosis was detected by flow cytometry: the apoptosis rate of the negative control group was 24 h 2.93% and 48 h 5.07%.The early apoptosis rate of sh SETD8 cells was significantly increased in 24 h 15.2%and 48 h 13.1%(P < 0.01).The expression of p53 protein was detected by Western blot method,and the expression of p53 in sh SETD8 cells was in high water(P < 0.01)compared with that in the negative control group.Conclusion:SET8 can be used as an indicator of tumor prognosis after surgery,and its high protein expression indicates poor prognosis of laryngeal cancer.SET8 gene affects the transformation of laryngeal cancer by regulating the proliferation,migration,invasion and apoptosis of laryngeal cancer cells.The expression of SET8 protein is closely related to SUVmax value,and SUVmax can be used as an effective indicator to predict tumor prognosis before surgery,and the high SUVmax value indicates poor prognosis of laryngeal cancer.
