Objective The aim of this research was to explore the effects of EA at ST 36 on gastric emptying and blood glucose in chimeric diabetic mice.Methods The bone marrow cells from male GFP-transgenic C57BL/6 mice were injected into the wild-type C57BL/6 mice with total-body irradiation via tail vein.The rate of chimerism was examined after one month.66 mice were used for subsequent experiments only when 80% or more of their blood cells had been replaced with GFP+cells.The mice were randomly divided into 6 groups: Control,diabetic(DM),bone marrow transplantation(BMT)plus diabetic(BMT+DM),BMT plus diabetic plus sham EA(BMT+DM+SEA),BMT plus diabetic plus low-frequency EA(BMT+DM+LEA),and BMT plus diabetic plus high-frequency EA(BMT+DM+HEA).The blood glucose of mice in each group was recorded before modeling,1 week after modeling,and 8 weeks after the model,respectively.At the end of the 8th week after the model,gastric emptying was detected by gavage with phenol red meal.Results(1)The average chimerism rate was approximately 90.0% in the chimeric mice.(2)No differences in blood glucose levels were found between the DM group and the BMT+DM group before modeling,1 week after modeling,and 8 weeks after the model,respectively.Blood glucose levels in the BMT+DM+SEA,BMT+DM+LEA,and BMT+ DM+HEA groups did not change compared with those in the BMT+DM group before modeling,1 week after modeling,and 8 weeks after the model,respectively.(3)Gastric emptying of the DM group was signifcantly delayed compared with that in the control group.The gastric emptying was significantly accelerated in the BMT+DM+LEA and BMT+DM+HEA groups compared with that in the BMT+DM group.Conclusion(1)The GFP+ chimeric mouse model is successfully established.(2)Bone marrow transplatation has no effect on blood glucose;EA at ST36 has no effect on blood glucose levels in chimeric diabetic mice.(3)EA can improve delayed gastric emptying in chimeric diabetic mice.Objective The aim of this study to explore the effect of EA at ST 36 on the ICC of bone marrow derivation in the stomach of diabetic mice.Methods c-Kit+GFP+ cells in the stomach were detected by immunofluorescence staining.Western blotting and q RT-PCR were employed to detect the m RNA expression and protein expression of GFP and c-Kit.Results(1)Numbers of c-Kit+GFP+cells among ICC-IM and ICC-MP were markedly elevated in the BMT+DM+LEA and BMT+DM+HEA groups compared with those in the BMT+DM and BMT+DM+SEA groups.(2)c-Kit protein levels were significantly lower in the DM group than those in the control group.c-Kit protein levels were significantly increased in the BMT+DM+LEA and BMT+DM+HEA groups compared with those in the BMT+DM group.(3)The m RNA and protein expression level of GFP were significantly increased in the BMT+DM+LEA and BMT+DM+HEA groups compared with those in the BMT+DM group.Conclusion Both low-and high-frequency EA at ST36 can increase the number of bone marrow-derived ICC in the stomach of diabetic mice.Objective To investigate whether SDF-1/CXCR4 and m SCF/Kit-ETV1 signaling pathways are involved in the process of EA increasing bone marrow-derived ICC in the stomach of chimeric diabetic mice.Methods The protein expression of SDF-1,CXCR4,m SCF,p ERK and ETV1 in gastric tissues were determined by Western blot.QRT-PCR was employed to detect the m RNA expression of SDF-1,CXCR4,m SCF,p ERK and ETV1 in gastric tissues.SDF-1 protein levels in the serum were detected by ELISA.Results(1)A distinct up-regulation of SDF-1 and CXCR4 at protein and m RNA level was observed in the BMT+DM+LEA and BMT+DM+HEA groups compared with those in the BMT+DM group.(2)SDF-1 protein levels in the sera were markedly increased in the BMT+DM+LEA and BMT+DM+HEA groups compared with those in the BMT+DM group.(3)The protein and m RNA expression levels of m SCF,p ERK and ETV1 in the gastric tissues were significantly lower in the DM group than those in the control group.m SCF,p ERK and ETV1 expression levels were significantly increased in the BMT+DM+LEA and BMT+DM+HEA groups compared with those in the BMT+DM group.Conclusion Both low-and high-frequency EA at ST36 increase the number of bone marrow-derived ICC in the stomach of diabetic mice via the SDF-1/CXCR4 and m SCF/c-Kit-ETV1 signaling pathways.
