Purpose: Currently,experimental studies have confirmed that many miRNA and target genes have inhibitory information pathways,while the target genes are mostly cancer-related genes.PDCD10 is identified as an apoptosis?related gene that play an important role in regulating cell apoptosis.PDCD10 in non-small cell lung cancer,high expression in poorly differentiated lung cancer is especially.Some studies have confirmed in prostate cancer miRNA-103 to inhibit cancer growth by inhibiting the expression of PDCD10.To study whether miRNA-103 exerts inhibitory effects on tumor by targeting PDCD10,we observe the effect of miRNA-103 overexpression or PDCD10 gene silencing on the proliferation and invasion of non small cell lung cancer.The purpose of this study was to investigate the miRNA-103 and expression of PDCD10 in non-small cell lung cancer.Methods: Real-time quantitative reverse PCR was used to express detection of mi R-103 and PDCD10 m RNA in 32 pair of two types of lung cancer and normal lung tissue.PDCD10 is one of the target genes by using a number of high throughput databases to screen the target genes of miRNA-103.Negative control mimic(NC)?miRNA-103 inhibitor?pc DNA3.1 PDCD10 or negative control siRNA(pc DNA3.1)were transfected into A549 cells respectively,and the expression of PDCD10 was detected.mi R-103 decreased the invasion and migration ability in A549 cells,as evidenced by Transwell and Wound healing results.Downregualtion of mi R-103 significantly reduced programmed cell death 10(PDCD10)level.We found a significant decrease to the relative luciferase activity of the reporter gene in A549 cells co-transfected with mi R-103 mimic and p GL3-PDCD10 WT 3'-UTR,but not pGL3-PDCD10 mut 3'-UTR.We showed that overexpression of PDCD10 significantly inhibited mi R-103-induced inhibition of cell proliferation,increased of apoptosis and decrease of invasion and migration in A549 cells.Results: In human non-small cell lung cancer cells,low expression of PDCD10 and miRNA-103 high expression was significantly correlated.The increased expression of PDCD10 protein in non-small cell lung cancer.Inhibition of miRNA-103 expression by transfection of mimics PDCD10.miRNA-103 mimics inhibit containing the luciferase activity of WT PDCD10-3'UTR plasmid and effects are not containing the luciferase activity of mut PDCD10-3'UTR plasmid,this shows that miRNA-103 direct role in PDCD10-3'UTR prediction of target sites.Enhanced expression of miRNA-103 or siRNA mediated PDCD10 gene silencing significantly decreased cell proliferation and invasion of lung cancer.Conclusions: miRNA-103 Negative regulates the translation products of m RNA PDCD10.Enhanced miRNA-103 expression or siRNA mediated PDCD10 gene silencing can significantly reduce the proliferation and invasion of lung cancer cells.
