| Purpose:To explore the mechanism of effect of Phlegm dissolving,stasis dispering and stagnation unblocking formula on spontaneous movement and learning and memory activity in schizophrenic model rats.To discuss Phlegm dissolving stasis dispering and stagnation unblocking formula through regulating GJIC function and inhibiting oxidative stress reaction,neuroprotective effects,mechanisms for treating schizophrenia Methods:The model group,control group and treatment group are selected to use MK-801(0.1 mg/mL)intraperitoneal injection,to establish a rat model of Glu in schizophrenia.When model is successfully prepared,control group with Risperidone suspension(0.036mg/kg),the treatment group is treated with Phlegm dissolving,stasis dispering and stagnation unblocking formula(9.9g/kg).The first,second,third and fourth weeks of gavage,do the test of assessment of the open field and Morris water maze experiment.The contents of PKC,p-Cx43 and p38MAPK in serum of rats with schizophrenia are detected by Elisa.Western Blot and Real-Time PCR are used to detect the protein and gene expression levels of NADPH oxidase subunits p22 and p47 in rat hippocampal tissues of rats with schizophrenia.TUNEL method is used to detect the apoptosis of nerve cells in hippocampus of rats,Western Blot and Real-Time PCR are used to detect c-fos protein and gene expression in hippocampal tissues of rats with schizophrenia,immunofluorescence assay is used to detect the expression of s100-beta protein in rat hippocampal tissue.Results:1.Effect of Phlegm dissolving,stasis dispering and stagnation unblocking formula on spontaneous activity and learning and memory activity in rats with schizophrenia model;after doing the drug intervention,comparing with control group,the total exercise distance of the treatment group(day 21,day 28)and the total number of squares crossed(day 21,day 28)was significantly increased,the number of leg lifts is similar to that of the grooming,escaped the incubation period was significantly reduced(week 3),the stay time in the original platform quadrant increased(week 3,week 4),the number crossing the original platform in the 60s is similar.2.Effects of Phlegm dissolving,stasis dispering and stagnation unblocking formula on the effects of GJIC function in schizophrenia model rats:after doing the drug intervention,comparing with control group,the content of PKC in rat serum is similar to the treatment group,contents of p-cx43 and p38MAPK was significant decreased.3.Effect of Phlegm dissolving,stasis dispering and stagnation unblocking formula on the content of NADPH oxidase in hippocampus of rats:after doing drug intervention,comparing with control group,the expression of NADPH oxidase subunit p22 and p47 in hippocampal tissue of rats in the treatment group was significantly decreased.4.Effect of Phlegm dissolving,stasis dispering and stagnation unblocking formula on neuroprotective mechanism in rats with schizophrenia model:after doing drug intervention,comparing with control group,apoptosis rate of nerve cells in hippocampal tissue of rats in treatment group,c-fos protein and gene expression and the expression of s100-complement positive cells was significant decreased.Conclusion:1.Phlegm dissolving,stasis dispering and stagnation unblocking formula can increase voluntary activity of MK-801 induced schizophrenic model rats,anxiety decreased,learning and spatial memory improved,and it works better than Risperidone.2.Phlegm dissolving,stasis dispering and stagnation unblocking formula can reduce the content of PKC,p-Cx43 and p3 8MAPK in the serum of MK-801 induced schizophrenia model,then adjust the GJIC function,and it works better than Risperidone.3.The protein and gene expression levels of NADPH oxidase subunit p22 and p47 in the hippocampus of rat hippocampal tissues induced by MK-801are reduced by using Phlegm dissolving,stasis dispering and stagnation unblocking formula,improve its oxidative stress state,and it works better than Risperidone.4.The apoptosis rate of nerve cells in schizophrenia model rats induced by mk-801,the protein and gene expression of c-fos in hippocampal tissue,the positive expression of s100-complement nerve cells can be down-regulated by Phlegm dissolving,stasis dispering and stagnation unblocking formula.It can protect brain nerve cells and glial cells,improve brain function,and it works better than Risperidone. |
