Effect Of Qinggan Huadu Zhuyu Decoction On Patients With Liver Cancer After Interventional Therapy And Effects Of Ophiopogon Japonicus Saponin B On JAK2/STAT3 Pathway

Objective Observe the clinical curative effect of Qinggan Huadu Zhuyu Decoction in the treatment of patients with hepatocellular carcinoma after interventional intervention;To investigate the effect of Ophiopogon saponin B on human hepatocellular carcinoma MHCC97-H cells,evaluate cell viability by CCK-8 assay,and detect cell cycle by flow cytometry And cell apoptosis,detection of JAK2/STAT3 signaling pathway to explore the mechanism of Ophiopogon saponin B on human liver cancer MHCC97-H cells.Methods Clinical research:86 cases of hepatocellular carcinoma patients identified by traditional Chinese medicine as syndromes of dampness,heat and phlegm toxicity were randomly divided into two groups.The middle treatment group of Qinggan Huadu Zhuyu Decoction(42 cases)and Huaier granules control group(41 cases)were treated.The total course of treatment in the group and the control group was 3 months or more.The clinical curative effect of the patients was observed.The major changes in cancer size,TCM symptoms,HBV-DNA,AFP,survival rate,and drug safety were observed.Experimental study:Ophiopogon B(Op-B)is an active ingredient extracted from Ophiopogon japonicus(Op-B)from Qinggan Huadu Zhuyu Decoction.It is commonly used to treat cancer.The aim of the study was to investigate the effect of Op-B on human hepatoma MHCC97-H cells.Cell viability was assessed by the CCK-8 assay.Flow cytometry was used to detect cell cycle and apoptosis,respectively.Matrigel invasion experiments were performed to detect the invasion of Op-B induced MHCC97-H cells.Finally,the mechanism was studied using real-time quantitative polymerase chain reaction(RT-qPCR)and Western blotting.Results Clinical part:Qinggan Hudu Zhuyu Decoction can significantly improve the treatment of liver cancer after interventional treatment of hypochondriac pain,bloating,jaundice,fatigue,poor appetite,eat less,urinary symptoms,improve hypochondriac pain,bloating,jaundice,fatigue,poor appetite and less food In terms of symptoms,Qinggan Huadu Zhuyu Decoction is significantly better than the Huaier granule group.Qinggan Huadu Zhuyu Decoction can improve the patients'AST,ALT,total bilirubin,albumin level,alpha-fetoprotein level,HBV-DNA,prothrombin time significantly higher than that of Huaier granule group.Tang can significantly reduce the level of CD4+?CD8+?CD4+/CD8+and NK damage caused by interventional therapy of liver cancer,and is beneficial to the maintenance of the patient's immune function;Qinggan Huadu Zhuyu Decoction can significantly prolong the patient's risk of liver cancer after interventional treatment.Lifetime,improve their quality of life.Liver-clearing,poisoning and removing blood stasis have no significant effect on renal function.There were no obvious adverse reactions in patients with hepatocellular carcinoma,hepatectomy,liver transplantation,and liver cancer.The experimental results show that we detected the JAK2/STAT3 signaling pathway and found that Op-B inhibits the phosphorylation of JAK2 and STAT3.Real-time quantitative polymerase chain reaction and immunoblot results showed that Op-B can increase the mRNA and protein expression levels of caspase-3 and Bax,and at the same time reduce Bcl-2 and PCNA.MMP-2,VEGF and VEGFR2 expression levels.In addition,IL-6 treatment not only activates STAT3,but also inhibits the expression of OPNA-induced PCNA,Bcl-2,MMP-2,VEGF and VEGFR2,and increases the expression of Bax and Caspase-3.Therefore,inhibition of JAK2/STAT3 signaling pathway may be related to Op-B-mediated MHCC97-H cell viability and invasiveness.Conclusion The curative effect of QingGanJiangPi and PingTiaoGanPi method Sequential treating ulcerative colitis patients is better than that of 5-ASA,especially in improving TCM syndrome and intestinal mucous membrane.According to the experimental study,we conjecture that D-limonene reduced MMP-2 and-9 mRNA expression levels via regulation of the iNOS,COX-2,PGE2,TGF-? and ERK1/2 signaling pathways in a UC rat model,indicating its potential antioxidant and anti-inflammatory properties.Conclusion Qinggan Huadu Zhuyu Decoction can improve the survival rate of patients after liver cancer intervention and prolong the survival time of patients,improve the KPS score of patients after liver cancer intervention,improve the liver function of patients,reduce the level of hepatitis B virus and alpha fetoprotein in patients.,Ophiopogon japonicum saponin B through the inhibition of JAK2/STAT3 pathway to achieve liver cancer MHCC97-H cell activity,invasion and proliferation.
