Part One Portal vein ligation promotes liver regeneration in rats with obstructive jaundiceObjective:To build the rat model of obstructive jaundice and investigate the effect of port vein ligation on liver regeneration of non-ligation part in the rats of obstructive jaundice.Methods:Ninety male Sprague-Dawley rats were randomly divided into three groups:control group of sham operation(C-Sham)?control group of obstructive jaundice(C-OJ)and selective port vein ligation group(PVL).The bile ducts of left and median lobes and portal vein were only dissociated in the rats of C-sham group;The bile ducts of left and median lobes were cut off in the rats of C-OJ group to build the rat model of obstructive jaundice;Besides the above steps of C-OJ group,the portal vein of left and median lobes were ligated.At each testing time point,samples were taken to test the ratio of non-ligation liver part?the expression of KI-67?serum total bilirubin(TBIL)?direct bilirubin(DBIL)?albumin(ALB)?alanine aminotransferase(ALT)and aspartate aminotransferase(AST);The part of ligation and non-ligation liver were distinctively stained with hematoxylin-eosin(H-E)to observe the histological changes of liver cells.Results:the ratio of non-ligation liver part and the expression of KI-67 in non-ligation liver part in PVL group were higher than that in C-OJ group and C-sham group(P<0.05);At 48 hours?72 hours?120 hours and 168 hours after operation,the ratio of non-ligation liver part and the expression of KI-67 in non-ligation liver part in C-OJ group were higher than that in C-sham group(P<0.05).The serum TBIL and DBIL in C-OJ group and PVL group were higher than that in C-sham group(P<0.05);At 36 hours after operation,the serum TBIL and DBIL in PVL group were distinctively(8.19±2.16)umol/L?(7.12±2.25)umol/L,higher than(4.34± 1.56)umol/L?(2.85±1.40)umol/L in C-OJ group.At 48 hours after operation,the serum ALB in PVL group was(23.75±3.16)g/L,lower than(31.25±2.03)g/L in C-sham group and(30.32±3.35)g/L in C-OJ group(P<0.05).At 36 hours after operation,the serum ALT and ADT in PVL group and C-OJ group were the highest,and the serum ALT and AST in PVL group were(356.53±74.36)U/L and(843.76±156.43)U/L,higher than(146.62±50.24)U/L and(501.87±142.54)U/L in C-OJ group.There were significant differences between the two groups(P<0.05);At 48 hours?120 hours and 168 hours after operation,there were no significant differences of serum ALT between PVL group and C-OJ group(P>0.05);At 120 hours and 168 hours after operation,there were no significant differences of serum AST between PVL group and C-OJ group(P>0.05).In PVL group,liver cells became smaller and there were visible necrosis areas in the ligation part of liver,however,liver cells in the non-ligation part became bigger and showed more hyperchromatic nuclei and nuclear division.Conclusion:Ligating 70%bile duct of rats' liver could stably build rats model of obstructive jaundice;portal vein ligation could safely and effectively promote liver regeneration of non-ligation part.Part twoBasic study of signal transduction pathway for liver regeneration in rats of obstructive jaundice promoted by port vein ligationObjective:To study the signal transduction pathway for liver regeneration in rats of obstructive jaundice promoted by portal vein ligation.Methods:According to certain time point,rats' serum,ligation part of liver and non-ligation part of liver were get to test the expression of TNF-? and IL-6 by ELISA;The expression of HIF-1??P65 and P50 of NF-?B were tested by western-blot 48 hours after operation,the nuclear P50 expression of liver cells in the non-ligation part was tested by immunohistochemistry.Results:After operation,the expression of TNF-? and IL-6 in C-OJ group and PVL group increased,and the expression reached the highest at 48 hours after operation.The expression of TNF-? in C-OJ group was higher than that in C-sham group except at 168 hours after operation;The expressions of TNF-? on each time point in PVL group were higher than that in C-sham group and C-OJ group;The expressions of IL-6 on each time point in C-OJ group and PVL group were higher than C-sham group;The expressions of IL-6 on each time point in PVL group were higher than C-sham group and C-OJ group.The expression of HIF-la in C-sham group and C-OJ group wasn't apparent;The HIF-la expression in liver cells of ligation part increased,however,the expression in liver cells of non-ligation part wasn't apparent.The expression of P65 and P50 increased significantly in the ligation and non-ligation part of C-OJ group and PVL group;At 48 hours after operation,the expression of nuclear P50 in the non-ligation part of PVL group was higher than that of C-sham group.Conclusion:Portal vein ligation could promote liver regeneration of non-ligation part by highly expressing TNF-? induced by HIF-la,and then by involving the signal transduction pathway of NF-?B and IL-6.
