| Objectives1.To analyse the experience of medicinal guides in herbs to treat the fundus diseases from Professor Tang Youzhi;To review and investegat the clinical cases of medicinal guides in herbs to treat the fundus diseases,and summarize my tutor's experience.2.To observe the contribution of loganin and morroniside of Liuwei Dihuang pills affected by adding different medicinal guide herbs through modern research technology.3.To observe the expression of claudin-5 protein in blood-retina barriers of SD rats induced by NaI02 through molecular biology techniques.Methods1.The experience of Proffessor Tang were learned by reading books and anlysing clinical record.The patients during 2013.3-2014.9 in my hospital were reviewed to observed the clinical case of medicinal guides in herbs to treat the fundus diseases.2.The effect of borneol,bupleurum and Radix Saposhnikoviae on the distribution and metabolism of components of LwDH pill in different eye organization effects:rabbits were divided into 5 group randomly;the liquid of distilled water,LwDH pill,LwDH pill + borneol,LwDH pill + Radix Saposhnikoviae and LwDH pill + bupleurum were irrigated respectively;to observe the content of loganin and morroniside of liuwei dihuang pill in aqueous humor,Vitreous body and retina of rabbits by LC-MS method before medication(0h)and medication after 0.25h,0.50h,0.75h,1h,2h,4h,8h;To investigate whether the medicinal guide promoted the gathering of drug components targeting in eyes through the mechanism as carrier.3.SD rats were induced to retinal injury model by iodate sodium as research object,to observate the effect of drugs in pathology state.Medication were given when pathology change occurred in retina appeared obviously after seventh days.To observe the expression volume and location of the blood-retinal barrier composition protein Claudin-5 of each group at different time point before irrigation and irrigated after 30min,1h,2H,4H,and 8h by immunohistochemistry and Western blotting.Result1.To summarize the clinical experience of Proffesor Tangyouzhi from the perspective of dialectics,compatibility and characteristics;To explore the academic thought of Professor Tang to use the dispelling wind and resuscitation herbs with different formulae of clinical application and research.Statistics of retrospective cases showed that there were 47 cases of patient medicated by dispelling wind and resuscitation herbs,in 178 cases;In which there were 38.30%medicated by Radix Saposhnikoviae and 51.06%by bupleurum.2.Loganin and morroniside can be detected in the plasma of rabbits;each component distributed in the plasma quickly.From the trends of graphs,the change of concentration in each group apeared similar;the peak appeared after 0.25h?0.75h,and cleared up after 8h basically.The plasma concentration of morroniside and loganin in borneol group were higher than the other groups after 0.25h and 1h;there were significant differences(P<0.05);The concentration in aqueous humor of morroniside and loganin in borneol group were higher than the other groups after 0.25h?0.75h;there were significant differences(P<0.05);The concentration in Vitreous body of morroniside in borneol group were higher than the other groups after 0.25h?0.75h;there were significant differences(P<0.05);The concentration in retina of morroniside and loganin in borneol group were higher than the other groups after 0.25h;there were significant differences(P<0.05).3.IHC:claudin-5 mainly express in the neurosensory retinal microvascular endothelial cells and the RPE layer;the distributions of claudin-5 are continuously along the neurosensory retinal microvascular endothelial cells and the RPE layer,which in normal group,Oh group,distilled water group,and LwDH group;the distribution of borneol group,and Radix Saposhnikoviae group are obviously less than distilled water group and LwDH group irrigated after 0.5h and 1h.Western blot:there was no significant difference of claudin-5 protein expression in 0 hours group(F=1.684,P=0.247).Irrigated after 0.5h,the largest decrease of claudin-5 protein expression volume was Borneol group;irrigated after 1h,the largest decrease of claudin-5 protein expression volume was RS group;irrigated after 2h,the largest increase of claudin-5 protein expression volume was distilled water group;between the 6 time points,the expression of claudin-5 protein irritated after 0.5h,1h,2h and 4h are lower than Oh and 8h,which were significant different(P<0.05);there was no statistically significant difference between 8h and 0h(P>0.05).Conclusion1.Dispelling wind herbs were combined with tonifying drugs in the clinical treatment of ocular fundus diseases in most of the prescriptions.Professor Tang Youzhi likes to use bupleurum or Radix Saposhnikoviae combined with tonifying liver and kidney drugs in prescriptions.In clinic and research,the recruition herb boneol was always combined with topical ophthalmic preparations.2.Borneol can promote the evolution of effective components of LwDH in plasma with similarity and differences of morroniside and loganin,which indicated borneol may enhance the absorption of oral prescription in eyes by affecting the plasma-drug concentration.Borneol can enhance the concentration of components of LwDH in eye organisms,which indicated further that it may enhance the absorption of oral prescription in eyes by affect the capability through BRB.3.Borneol and Radix Saposhnikoviae may affect the permeability of BRB by suppress the expression of Claudin-5 to promote the eye-targeted treatment of oral prescription.This course was transient and reversible. |
