Expression Profile Of TSEG2,IZUMO1,SYCP3,and LHB Genes In Different Developmental Stages Of Yak Testis

This research was planned to compare the expression profile of TSEG2,IZUMO1,SYCP3 and LHB genes in Datong yak during different developmental stages.The expression patterns of these genes and their proteins were explored by using bioinformatics,qPCR,semi-quantitative PCR,western blot,and through Immunohistochemistry.Our observation of semi-quantitative PCR and qPCR indicates that expression of TSEG2,IZUMO1,SYCP3 and LHB genes were significantly unregulated in the testicles of Yak as compared to other tissues.In addition to that immunohistochemistry results showed the higher expression of IZUMO1;SYCP3 and LHB proteins in the testicles of six years followed by 30,18 and 6months respectively.Additionally,western blot examination indicated that the molecular weight of IZUMO1,SYCP3,TSEG2 and LHB proteins were 60-kDa,34-kDa,90-kDa and 16 kDa in testicles of Yak.Histological investigation of testicles is necessary for understanding its growth,sex development and problems of infertility.Hence,we did hematoxylin and eosin staining of Yak testis during different developmental ages.Our findings exposed that the spermatogenic cells and their nuclei diameters were significantly longer in the testicles at 6 years as compared to 18 and 6 and 8 months.Whereas no any significant difference was observed between six years and 30 months.Additionally,HE staining findings exposed that there was no any significant difference in morphological observation of Datong Yak testes at different ages.Though,thickness of epithelium,volume density of seminiferous epithelium and seminiferous tubules,and cross sectional area increased steadily when animal going towards advanced stage.The high expression of TSEG2,IZUMO1,SYCP3 and LHB in testicles directed that these genes may be significant for fertility and in the growth of gonads.
