“One Belt One Road” and “Sanjiangyuan Ecological Protection” are national strategic plans,and Qinghai Province is the key area.With the initiation of the national strategy,movement of human and products will increase ticks and tick-borne pathogens endangering the health of the residents and domestic animals.Studies on species diversity of ticks and gene polymorphism of tick-borne pathogen,and the risk factors for assessing the impact on ecological envirment and public health security.It is profound significance,which will provide results of this study for control of ticks and tick borne diseases in Qinghai Province and neighbor countries,and to our knowledge of ticks and tick borne diseases in depth.A total of 1104 questing adult ticks were collected from vegetation on 22 counties of Qinghai Province by flagging and trapping method.The species of ticks were identified through comprehensive morphology and based on the gene of 16 S rRNA,COI,ITS2 analysis to identify and it has been demonstrate that there were 454 Haemaphysalis qinghaiensis,2 H.tibetensis,42 H.danieli,263 Dermacentor abaensis,246 D.nuttalli,94 D.silvarum,and 3 Ixodes crenulatus.Pathogens were detected by PCR and RLB in this study.The results of PCR showed that the infection rates were 3.1%,11.1%,5.6%,4.5%,44.7%,5.6%,22.9% and 1.6% for Anaplasma phagocytophilum,A.bovis,A.ovis,A.capra,SFG rickettsiae,Borrelia.burgdorferi sensu lato,Piroplasm and Colpodella spp.respectively.A.marginale was not detected in any of samples.In phylogenetic analysis,4 differernt sequences identities to A.ovis of MSP4 gene,5 to A.bovis and 3 to A.phagocytophilum of 16 S rRNA gene,2 to B.burgdorferi sensu lato of fla gene,3 to Babesia and 5 to Theileria of 18 SrRNA gene,2 to A.capra of gltA,16 S rRNA,groEL genes,3 to Colpodella spp.of 18 SrRNA gene.The results of RLB showed that 4 species and 4 types of mixed infections of B.burgdorferi sensu lato were recognized,and 6 species and 3 types of mixed infections of Piroplasm were also found in ticks from Qinghai Province.And then using bioinformatics software in data of pathogens detected for assess the risk of epidemic disease,and the factors were analyzed affecting the high incidence of epidemic disease.The MSP1 a amino acid repeats of A.ovis were analyzed gene polymorphism from ticks in Qinghai Province,and from sheeps in 24 counties from 12 provinces of China.34 partial MSP1 a gene sequences contained the repeat sequences were obtained from A.ovis-positive samples of ticks,20 different types of MSP1 a repeats were identified and 18 genotypes based on the organization of different amino acid repeats of A.ovis in ticks,and 44 partial MSP1 a gene sequences contained the repeat sequences were obtained from A.ovis-positive samples of sheeps.24 different types of MSP1 a repeats were identified and 19 genotypes based on the organization of different amino acid repeats of A.ovis in sheeps.
