| Nicotiana tabacum L.is a plant belonging to the Nicotiana L(Solanaceae),in which sun/air-cured tobacco becomes an indispensable raw material for coordinating the taste quality of cigarettes based on its unique modulation.Pueraria lobata(Wild.)Ohwi is dry root of Leguminous sp.plant.As an additive of tobacco filter tip,its particles can improve the smoking comfort of tobacco products.Phlomis betonicoides is a years herbaceous plant of Phlomis Linn.(Labiatae),whose ethanol extract can be used as the additive of cigarette filter tip to increase the sweet and moist smoke of cigarette products.All three were used for traditional cigarette smoking quality improvement and new product development in the tobacco industry.The chemical constituents of the three plants were studied in this paper,which would provide a basis for the rational exploitation and comprehensive utilization of these three plants.In the first chapter,the chemical components of sun/air-cured tobacco and their bioactivity research status were reviewed,the compounds separated and identified from sun/air-cured tobacco were emphatically introduced,and the anti-tobacco mosaic virus and cytotoxic activities of part of the compounds were mentioned.In the second chapter,the chemical compositions of sun/air-cured tobacco from Weishan,Yunnan and their bioactivities were studied.76 compounds,including 17 new ones 1-17,were isolated and identified,containing 3 isoindolinone compounds(1-3),6 phenylpropane compounds(4-7 and 9-10),2 furan carboxylic acids(11-12)and 6 isobenzofurans(8 and 13-17).Compounds 1-3,16-17 showed good anti-tobacco mosaic virus activities at the concentration of 20 μM,with inhibition rates of 43.8%,45.6%,52.7%,35.4%and 34.2%,respectively.It was higher than the positive control(ningnamamycin).The compounds 1-8,15-17 showed moderate cytotoxic activities against NB4,A549,SHSY5Y,PC3 and MCF7 in vitro,with the IC50 value range of 2.5-9.5 μM.In the third chapter,the chemical compositions of Pueraria lobata(Wild.)Ohwi and their bioactivities were studied.37 compounds,including 8 new isoflavones 1-8,were isolated and identified.Compounds 2,3,4,5 and 8 showed higher anti-tobacco mosaic virus activities than positive sample at the concentration of 20 μM,with inhibition rates of 34.2%,33.5%,36.8%,33.6%and 32.6%,respectively.The compounds 1-13 showed moderate cytotoxic activities against NB4,A549,SHSY5Y,PC3 and MCF7 in vitro,and the IC50 value range was 1.2-9.8 μM.In the forth chapter,the chemical constituents of Phlomis betonicoides and their bioactivities were studied.40 compounds including 3 new ones,containing 2 new isoflavones(1-2)and 1 new phenylpropane(3),were isolated and identified.Compounds 1 and 2 showed good antioxidant activities with IC50 values of 3.8 and 3.6μM,respectively.The compound 3 showed prominent antibacterial activity with MIC90 value of 58.4 μM for methicillin resistant Staphylococcus aureus(MRSA).This value of MIC90 was close to that of levofloxacin(with MIC90 value 52.8 μM).In this paper,153 compounds including 28 new compounds were isolated and identified,and a large number of compounds with certain biological activities were discussed.The chemical and bioactive researches of the three plants(Nicotiana tabacum L.,Pueraria lobata(Wild.)Ohwi,and Phlomis betonicoides)were enriched.At the same time,a study on the sensory evaluation of new compounds in cigarette was carried out.The scientific basis for the improvement of the quality of traditional cigarette smoke as well as the further exploitation and utilization of these three plants were provided. |
