| There are abundant chemical components in plants,some of which have the efficacy of immunoregulation,prevention and treatment of diseases,and can also be used in the development of health products,food,medicines and cosmetics.Functional foods have become a popular style of the healthy life due to their security,supplement nutrition and disease prevention.Therefore,the research and exploration of functional active components in plants is of great scientific significance for the development and utilization of plant resources at a multi-level level.The fruits of Swietenia macrophylla(S.macrophylla)King(Meliaceae),which is originated from the Solomon Islands in the South Pacific,and introduced in Yunnan,Hainan,Guangdong,Guangxi,Fujian and Taiwan in China.The health benefits of S.macrophylla fruits are greatly concerned,it can be used to improve immunity,and is good for preventing the diseases of liver,cardiovascular,diabetes,hypertension,hyperlipidemia,as well as treating high cholesterol and so on.Camellia oleifera(C.oleifera)flowers is the reproductive organ of plants,which is rich in active ingredients and is mainly used for cooling blood and hemostasis in the folk.They are widely cultivated as oil-bearing crop in the south of China,which are distributed in Sichuan,Jiangsu,Zhejiang,Fujian,Taiwan,Jiangxi,Hunan,Hubei,Guangdong,Guangxi,Yunnan,et al.For further research on these two plants,our study was focused on the isolation of major components of S.macrophylla fruits and C.oleifera flowers,we evaluated the pharmacological activities of the components of S.macrophylla fruits,established the analytical methods for the flavonoids of C.oleifera flowers which acquired satisfied result.The results were as shown below:1.The extraction,separation and purification of the ingredients from the fruits of Swietenia macrophyllaThe S.macrophylla fruits were reflux-extracted with methanol,and the crude extract was dissolved in distilled water and solvent-partitioned with EtOAc,then affording EtOAc-soluble Fr.After a preliminary separation,Fr F and Fr G parts which were the main components were obtained,then analgesic and anti-inflammatory activity tests were carried out with the mice model.It was proved that the crude extract had activity.Then with the application of various methods of separation and purification,such as silica gel column chromatography,ODS column chromatography,MCI column chromatography,preparative HPLC,recrystallization and so on,a total of33 compounds were isolated and purified.2.Structural identification of the ingredients of the fruits of Swietenia macrophyllaThe traditional analysis techniques,such as MS,IR,UV,1D and 2D NMR,etc,were used to identify the isolated compounds.A series of limonoids compounds from the seed of S.macrophylla were identified,including 8 new limonoids compounds,as shown below:3-O-Propionylproceranolide,Swietemacrolide A,6-O-Acetylswietenin B,SwietemacrolideB,6-Deoxyswietemahonin A,SwietemacrolideE,Swietemacrolide C and Swietemacrolide D.3.Study on the functional activity of the ingredients from the fruits of Swietenia macrophyllaH2O2-induced damage process in the HUVECs was similar to the peroxide environment that occurs in the old people with high proportion.The limonoids compounds are the main ingredients from the EtOAc-soluble fraction of sky fruits extracts,and the structures of some of them are rich in oxygen functional group.The activity test of anti-oxidative damage on some limonoids compounds was carried out,the results revealed that swietemacrolide C(7)has remarkable inhibition effect against H2O2-induced dysfunction of HUVECs,which might be used to decrease excessive active oxygen in the human body and protect people from the health risk of cerebrovascular diseases.4.The extraction,separation and purification of the ingredients of Camellia oleifera flowersThe C.oleifera(Theaceae)flowers resource were abundant,the ingredients of which were extracted by infusion method with dichloromethane and 95%ethanol and followed by the process of isolation and purification.The traditional isolation methods of column chromatography were used,such as MCI,MPLC(medium pressure preparative liquid chromatography),ODS,sephadex LH-20,polyamide,silica gel and recrystallization,etc.Finally,19 compounds from the extracts were obtained.5.Structural identification of the ingredients of Camellia oleifera flowersThe obtained compounds from C.oleifera flowers were identified by the combination of a variety of analysis technique(1H-NMR、13C-NMR、LC-MS)and the literatures of the reported compounds.The identified compounds contain flavonoids,phenolic acids and so on,of which flavonoids are the main components.6.Deep eutectic solvents used as extraction solvent for the determination of flavonoids from Camellia oleifera flowers by high-performance liquid chromatographyA method for simultaneous determination of four different polar flavonoids from C.oleifera flowers was established for the first time.With the application of ultrasound-assisted extraction,deep eutectic solvents(DESs)was used as extracting solvent.The related experimental extraction factors were investigated by Box-Behnken design combined with response surface methodology,and the content of flavonoids from C.oleifera flowers was determined by HPLC-UV.The developed method was simple,quick and environment-friendly,displayed high precision and stability from the aspects of recovery(75.26%to 103.99%),RSDs of intra-day(<3.96%)and inter-day(<5.80%),and was suitable for the routine analysis of C.oleifera flowers. |
