By modifying one natural long noncoding RNA(lncRNA),the first universal protein expression enhancement technology,which regulates mRNA translation at post-transcriptional and pre-translational level by recruiting polysome to target mRNA to accelerate translation without affect the mRNA level,was developed,and termed RNAe(RNAe enhancement).RNAe has a length of 1.3k nucleotide and simple construct,which mainly contains pairing segment,inverted SINEB2 element,and partial ALU.RNAe is easy to be constructed,and has high specific targeting,and universality in mammalian cells.We showed the high applicable value of RNAe in antibody production and high throughput screenning for functional genes by verifying its enhancement o n antibody-10E8 and good effect in screening of genes about cell proliferation.We also scanned 17 k RNAe-like lncRNA from mouse genome according to the conformation of RNAe,and confirmed 4/8 at last,which revealed that RNAe-like lncRNAs are a kind of now ones,and it is very meaningful to study them.Mechanism study of RNAe is needed during its development and application.The pairing segment and SINEB2 segment(especially the loops in the secondary structure)are the necessary and sufficient condition of RNAe.Besides,we confirmed the direct binding of RNAe with ILF3 or eIF4A1,although their function to RNAe is still unknown.
