In order to understand the biological changes in the process of ecological restoration in the Sanjiangyuan,the soil of the forage experimental plot and the third larva of H.Sinense were collected in Sanjiangyuan region.The forage in the experimental field is used to restore the “heitutan” grassland in Sanjiangyuan region.The DNA of all samples were sequenced by the technology of High-throughput and SMAT.The structure of soil microbial community and gene expression profile of larva the third larva of H.Sinense were observed.The results show:1.There was not significant difference between the mean values of the percentage value of optimized sequence number of the samples in two years(P > 0.05).The ratio of bacterial content in 2012 A and 2012 B was significantly lower than other samples(P < 0.03).The mean value difference was not significant between the samples of the forage field in two years(P > 0.05).2.There were 62724 ORF in DNA of 2013 D,accounting for 0.11% of the optimized sequence.Annotated 30715 ORF sequences in NR,684 in COG/KOG,and there were 18164 in KEGG,which are distributed in 69 metabolic pathways.Identification of species by Gene function and SSUrRNA homology,Proteobacteria?Actinobacteria and Acidobacteria were the dominant phylum.3.The genes of the gene expression profile of the third larva of H.Sinense,annotated in NR.There are 12 flies species(each of Musca domestica,Ceratitis capitata,Calliphora stygia and unknown species;8 Drosophila varieties.)were found.Among them,the number of annotated genes with Musca domestica is the highest(5375).There are 4898 ORF annotated in GO database.3207 in COG/KOG/NOG.There are 4456 in KEGG,which are distributed in 42 metabolic pathways.and only the glycolytic / glycolytic pathway was found,but there was no pathway related to aerobic oxidation metabolism.It may be suggested that the development and metabolism of larvae or pupal is relatively low for energy demand.In addition,this study also found 123 transcription factors of 26 classes.Among them,there are 44 transcription factors in zinc finger family,and many transcription factors are related to hypothermia adaptation.Conclusion: 1.It is proved by microbial community analysis that Pasture planting can restore the“heitutan” grassland ecology.Soil microbial community structure can be used as a microcosmic index for ecological restoration.Moreover,It is also confirmed that the connection is not obvious between the planting years of Poa alpigena in the experimental plot and the soil microbial Community structure in experimental plot.There is no obvious correlation between the planting years of Kentucky bluegrass and the soil microbial community structure in the experimental field.2.The functions of genes and transcription factors in the gene expression profiles of the third larvae of H.Sinense,has characteristics to tend to pupa and adapt to low temperature and low oxygen.3.The organisms in the area of Sanjiangyuan has obvious regional uniqueness.
