Effects Of High Fat Diet On Susceptibility To Colorectal Tumor And Intervention Of P-cymene

[Objective]To investigate the effect and mechanism of high-fat diet on 1,2-dimethylhydrazine(DMH)-induced colorectal tumor in Wistar rats.Interfering with p-cymene and its mechanism were studied.[Methods]Eighty wistar rats were randomly divided into 8 groups:normal diet group,normal diet + DMH group,normal diet + p-cymene group,normal diet + DMH + p-cymene group,high fat diet group,High fat diet + DMH group,high fat diet + p-cymene group,high fat diet + DMH + p-cymene group,10 rats in each group.The changes of serum lipid metabolism(total cholesterol,triglyceride,HDL,LDL,FABP4),inflammatory cytokines(IL-1,IL-6,COX-2,TNF-a,ADP and LEP),intestinal tumors,intestinal FABP4,TAOC,SOD,MDA and intestinal macrophages were analyzed on the status of high-fat diet,DMH induction and p-cymene.[Results]There are four cancerous nodules on four rats(40%)formed in normal diet +DMH group,while two cancerous nodules on two rats(22.2%)fored in normal diet + DMH +paraquat group formed.There are eight cancerous nodules on seven rats(70%)formed in the high-fat diet + DMH group,while eight cancerous nodules on the four rats(44.4%)formed in the hypercholesterolemic diet + DMH + paraquat group.High-fat diet resulted in the increase of serum cholesterol(P<0.05),the decrease of FABP4(P>0.05),the decrease of IL-1 and IL-6(P>0.05).The expressions of TAOC,SOD and MDA in intestinal tissue were increased,but the difference was not statistically significant(P>0.05).The proportion of M1 macrophages decreased and the proportion of M2 macrophages increased.Cypermethrin could reduce the level of COX-2(P<0.05)and increase the level of IL-1 and IL-6(P<0.05).[Conclusion]Hyperlipidemia increased the risk of colorectal cancer induced by DMH,and p-cymene could reduce the incidence of colorectal cancer induced by DMH.High-fat diet caused hyperlipidemia,the levels of serum IL-1 and IL-6 were decreased,the level of serum COX-2 was increased,the expressions of TAOC,SOD and MDA were increased in intestinal tissue.Cypermethrin could inhibit the expression of COX-2 and restore serum IL-1,IL-6 expression.According the point of TCM,High-fat diet cause "the status of chronic inflammatory"," the status of immune dysfunction ",and "the status of immune dysfunction" apears earlier than "the status of chronic inflammatory ",that is,"SHI" apears earlier than "RE" and "SHI"apears more serious than "RE";traditional Chinese medicine to "wake up" spleen can restore immune function.
