Objective: to detect the clinical significance of Cullin1 in patients with breast cancer and its biological effects on breast cancer cells.Methods: A total of 266 postoperative patients with breast cancer between 2008-2009 in our cancer center were included.The expression level of Cullin1 protein were analyzed by immunohistochemical.The effect of Cullin1 on proliferation of breast cancer cells was analyzed by cell proliferation assay.Transwell assay was used to analyze the effect of Cullin1 on invasive breast cancer cells.In addition,breast cancer cell lines stably overexpressing Cullin1 were constructed and transplanted tumor models were established in nude mice to observe the biological effects of Cullin1 on breast tumors.Results: Cullin1 expression was significantly associated with ER-status(P =0.01).Patients with Cullin1 high expression showed poored OS compared with patientswith Cullin1 low expression(P =0.034).Multivariate analysis confirmed that Cullin1(P=0.017)was an independent and significant prognostic factor for predicting the poor outcome of breast cancer patients.The proliferation rate of MDA-MB-231 and BT-549 cells with low expression of Cullin1 decreased significantly and the invasive ability also decreased.The volume and weight of tumor in nude mice inoculated with overexpressing Cullin1 inincreased significantly compared with those in the control group.Conclusions: These results provided evidence for the function of Cullin1 on the progression of breast cancer and suggested Cullin1 was a useful marker in prognostic evaluation for patients with breast cancer and maybe a therapeutic target in the future.
