Autophagy is a common immune response in cell,which is belonged to innate immune system.The autophagosome is composed of a bilayer membrane structure,which can degrade damaged organelles or some other unwanted components in the cell,after the degradation,the degraded small molecules can be reused by cells to maintain intracellular homeostasis.Autophagy related gene 8(Atg8)is the essential part for the autophagy membrane's elongation,in this progress,Atg8 need Atg4 to attach on the membrane.,Atg8 need Atg4 to be hydrated form,this form of Atg8 combine with PE to become the lapidated form,the lapidated Atg8 is important for the membrane's elongation,when the autophagosome is attached with lysosome,the Atg8 is cleaved by Atg4,based on this,the Atg4 is not only for Atg8 to transform to lapidated form,but also need Atg4 to cleave from the autophagosome.There are four homologous protein from Atg4 family,the effect of Atg4 protein on the Atg8 protein is not clear.In this study,we used human embryonic kidney cell(HEK 293T)to invest the function of Atg4 protein on Atg8 protein,and we have knock out different Atg4 homologous genes in HEK cell by CRISPR technology,then transfected the Atg4 gene into knock out cells by lentiviral transfection method in order to clarify the effect of Atg4 protein on Atg8 protein.The results showed that the Atg4 A protein played an important role in the lipolysis of LC3 B and GABARAPL1 which are belong to Atg8 family.Atg4 B protein played a role in the formation of soluble forms of LC3 B and GABARAPL1,but has the unique role for LC3 B protein.Atg4 C has a little function for LC3 B lipid form,Atg4 D has a little contribute for LC3B's lipidation,but for GABARAPL1's lipidation,it has a more significant role.Therefore,it can be concluded that the Atg4 protein has different effects on the transformation of the Atg8 protein from the pro-form to the soluble and lipidation form.In the following experiments,we can use different ways to test those results to confirm we got the correct answer,to make sure the development of autophagosome.From this study,we know the Atg4's function for autophagy,those results can guide us to study what's the relationship between the autophagy and cancer,especially for kidney cancer,which we can use autophagy to cure this disease,and to know how to treat for the immunological disease.
