Objectivewe want to explore potential mechanism of the expression of miRNA-214 in the femoral head with osteonecrosis and the change of activity of osteoblasts and osteoclasts. And the we prepare AAV-anti-miRNA-214 to regulate the activity of osteoblasts and osteoclasts to prevent the collapse of the femoral head with early stage osteonecrosis.Materials and MethodsThe sixteen femoral heads with early stage osteonecrosis were collected from patients with osteonecrosis. The samples were divided into different regions according to Micro-CT results, then osteoblasts and osteoclasts activity were analyzed by RT-PCR, Trap and ALP staining. We made adeno-associated virus carry anti-miRNA-214 and evaluated its ability of regulation of osteoblasts/osteoclasts activity. And then, we injected these gene medicines into rat femoral head with osteonecrosis and evaluated the ability of prevent the collapse of the femoral head with osteonecrosis.ResultsOsteoclast activity increased, osteoblast activity decreased and the expression of miRNA-214 increased in the necrisis region in the femoral head. We successful prepared the AAV-anti-miRNA-214, which can promote osteoblast activity,restrain osteoclast activity in vitro. Then we used these gene medicine to prevent the collapse of the femoral head in rats.ConclusionAAV- anti-miR-214 could promote osteoblast activity and restrain osteoclast activity, then effective prevent the collapse of the femoral head with early stage osteonecrosis.
