Exosomes are naturally occurring biological nanovesicles that are formed by the inward budding of multivesicular bodies.Recently, little is known about the interaction between melanoma exosomes and the microenvironment. In this research, exosomes were isolated, characterized and co-cultured with mesenchymal stromal cells(MSCs).It was found that exposure of MSCs to melanoma cell-derived exosomes enhanced MSC proliferative and migratory capability,as well as upregulation of expression of alpha-smooth muscle actin. All these effects were partly blocked by anti-TGF-?R antibody. It comes to the conclusion that melanoma cells may educate MSCs to modulate the microenvironment via TGF-?.Taken together, understanding the contributions of MSCs to tumour biology is of importance because they may result in new therapeutic or preventive paradigms. |
